Objective:This study aimed at examining the effects of verapamil on the peritoneal transport function and the peritoneal extracellular matrix in rats with peritoneal fibrosis induced by erythromycin and peritoneal dialysis solution.Methods:45 clean male SD rats were randomized into 3 groups(15 each) based on their weights.In Group A( the model group),erythromycin and peritoneal dialysis solution were injected to induce peritoneal fibrosis.In Group B( the intervention group),aside from the same treatment in Group A,intraperitoneal injections of verapamil were administered.In Group C(the control group),the rats were injected with normal saline at the concentration of 100m/kg.d.5 weeks after the model establishment,peritoneal equiliberation test was conducted,ultrafiltration volume and Mass Transfer of Glucose measured.In addtion,samples of parietal peritonea were given Masson's trichrome staining.Then MMP-9 in parietal peritonea was measured and the expression of fibronectin was assessed through immunehistochemistry;meanwhile the expression of TGF-?1 was accessed and its mRNA was measured by way of RT-PCR and Western-Blot.Results:After five weeks of treatment,UF volumes of the subjects in Group A and B were(-9.85±0.59) m L and(-4.36±0.41) mL respectively.MTG in Group A was(18.14±1.19)mmol/kg,in Group B,(14.24±0.67) mmol/kg.Peritoneal fibrosis scores were( 2.7±0.1)and(1.7±0.1) in Group A and B.In terms of MMP-9,Group A was(2.7±0.44)% while Group B was( 2.0±0.43)%.For FN,Group A was(2.1±0.32)%,Group B(1.7±0.31).Furthermore,in Group B,lower level of TGF-?1 expression and fewer mRNA,compared with those of Group A,were observed.To sum up,Group A and B have statistically significant differences in terms of all indicators,with P less than0.05.Conclusions:Our results indicate that verapamil can reduce TGF-?1 in the peritonea of rats with peritoneal fibrosis and curb the accumulation of extracellular matrix in the peritoneum,thus slowing down the process of peritoneal fibrosis.
