Objective: Generally used as a detoxifying drug by the people living in Guangxi,Caryopteridis Incanae Herba has the effect of dispelling wind cold,activating blood circulation to dissipate blood stasis,and eliminating dampness to relieve pain.So far,studies of Caryopteridis Incanae Herba mainly involve chemical ingredients,and no reports on its pharmacognostic identification and crude drug quality analysis have been available.The authenticity of crude drugs is the important guarantee of their safety and effectiveness as well as the basis for their rational development and application.In this study,morphological characters,microscopic structures and TLC features of Caryopteridis Incanae Herba were studied,water,ash and extract of its crude drug were done,the infrared characteristics and content of Caryopteridis Incanae Herba,and content of its volatile oil and total flavones was measured to establish systematic methods for pharmacognostic identification and quality analysis and provide references for quality control of crude drug of Caryopteridis Incanae Herba.Methods:(1)13 batches of crude drugs were subject to morphological and microscopic structure identification and TLC studies.(2)Water,ash and extract limit tests of the crude drugs were carried out as per Chinese Pharmacopoeia(2015).(3)infrared spectroscopy technology was used to research with spearmint infrared spectral characteristics of Caryopteridis Herba.(4)Using steam distillation to measure the content of volatile oil,we studied the change in the content of volatile oil of Caryopteridis Incanae Herba individuals collected before and after the flowering period and the full bloom stage and the effect of different drying methods on the content of volatile oil.(5)UV-Vis spectrophotometry was utilized to measure the content of total flavones;ultrasonic extraction of the total flavones was optimized by combining single factor experiment design with orthogonal experimental design.Results:(1)Plant and crude drug morphology,microscopic structures and TLC results were unique.(2)For different batches of crude drugs,the water content varied between 9.24%~11.82%,ash content between 2.34%~4.99%,acid insoluble ash content between 0.13%~1.48%,and 50% ethanol extract content between 14.65%~27.08%.(3)The Infrared spectroscopy of Caryopteridis Incanae Herba shows multiple characteristic absorption peaks,having a certain identify meaning.(4)The yield of volatile oil from Caryopteridis Incanae Herba was 0.05%(mL/g)~0.40%(mL/g),averaging 0.27%(mL/g).Caryopteridis Incanae Herba individuals gathered at the full bloom stage had a higher content of volatile oil than those collected before and after the flowering season,the extraction yields being 0.40%(mL/g),0.20%(mL/g)and 0.05%(mL/g);and the extraction yields of volatile oil for different drying methods were: dry in the shade>oven dry>dry in the sun.(5)The content of total flavones in Caryopteridis Incanae Herba crude drugs was 26.85mg/g~102.20mg/g.The optimal ultrasonic extraction process was as follows: extraction solvent,75% ethanol;sample to solvent ratio,1:40;soak period,30 min;ultrasonic extraction twice,45 min for each time,extraction power 300 W.Conclusion: The quality analysis method for Caryopteridis Incanae Herba established in this study is simple and fast,can be used as a theoretical basis for crude drug quality evaluation of Caryopteridis Incanae Herba.
