Objective:To establish the preparation process of the anti-rheumatism effective fractionfromtotalflavonoidsofHerbataxilli,pharmacodynamics observation of TFHT on adjuvant arthritis rats,explore the effective chemicals of TFHT and its distribution in AA rats and normal rats.Method:1.Effects of ethanol concentration,solid-liquid ratio,extraction time and temperature 4 factors on the yield of total flavonoids of Herba taxilli.On this basis,the orthogonal test was carried out to analyze the 4influencing factors,and the optimum extraction conditions were determined by the content of flavonoids in crude extract.The adsorption and desorption properties of the 4 resins were compared.To investigate the effects of the concentration of eluent ethanol and the flow rate of the effluent on the dynamic desorption of TFHT,the optimal desorption conditions were determined.2.Effects of TFHT on adjuvant arthritis rat model and its related substances:male SD rats were random Ly divided into normal,model,positive,blank,TFHT high,medium and low dose groups.All the other groups were injected with the same volume of Freund's complete adjuvant in addition to the normal saline injection group and 0.1m L group.After modeling for 8 days,from the beginning of the ninth day of stomach.Normal group,normal group,model group were given 0.5%sodium carboxymethyl cellulose solution by gavage,the positive control group were given Tripterygium wilfordii suspension?1mg/m L?,TFHT high?medium and low dose group were given TFHT?360mg/kg?,?180mg/kg??90mg/kg?,low dose of carboxymethyl fiber sodium solution.Once a day for 22 days.Fifteenth days,once every 7 days to measure the degree of toe swellingandajointscore.Gavagetwenty-seconddaysafterthe administration of all the rats weighing TFHT,high 2BV/h sample rate through AB-8 resin column?1BV=25m L?,after standing by large amounts of distilled water and impurity?proteins and polysaccharides?to the effluent 5BV and colorless,50%ethanol?v/v?,at a flowrateof2.5BV/helution,collectingtheeluent,thevacuum concentration and drying to obtain reddish brown powder,namely the purified TFHT.The content of TFHT was determined by UV and the content of total flavonoids was 56.73%,the yield was 5.5%.2.TFHT high and middle dose group can significantly improve the degree of swelling of the toes and the systemic symptoms of rats in adjuvant arthritis group;decrease the immune organ coefficient;decrease the levels of IL-6 and TNF-?,increased IL-10 content.Pathological sections showed ankle cartilage destruction was significantly reduced,exudate and pannus formation decreased or no.Pharmacodynamics experiments show that the total flavonoids of Herba taxilli has significant rheumatism and reversal of bone and joint damage.3.In the model of adjuvant arthritis model rats,the time of comparison was that quercitin was liver=kidney=heart=brain=spleen=small intestine>large intestine=lung>stomach>serum,Avicularin was Stomach>small intestine=brain>heart.The AUC0-t-t in the tissues were in the order of stomach>kidney>liver>spleen>small intestine>brain>heart>lung>large intestine>serum>Brain>heart,rutin only in the serum and stomach a small amount of distribution.In the normal group,the control time was compared waskidney=liver=stomach=smallintestine>brain>largeintestine>heart>splee n>lung>serum.Avicularinwaskidney>stomach>heart>smallintestine.The results showed that the quercetin was the number of the liver,the stomach,the small intestine,the intestine,the intestine,the heart,Small intestine The AUC0-t-t in the tissues were as follows:stomach>small intestine>kidney>liver>liver>small intestine>spleen>brain>heart>lung>large intestine>serum>Heart,rutin only a small amount of distribution in the stomach.Conclusion:Pharmacodynamic experimental results show that the total flavonoids of Herba Taxill has obvious effects of Anti-rheumatism,avicularin and quercetin through blood circulation,to have varying degrees of distribution in rat tissues.That avicularin and quercetin is the function of material flavonoids in the total flavonoids of Herba Taxill,Quercitrin,avicularin for rheumatism of the efficacy of flavonoids substances.Among them,quercetin is the total flavonoids of Herba Taxill meridian tropism and Anti-rheumatismeffectivechemicalsbasis.confirmedthatthetotal flavonoids of Herba Taxill is the effective parts of anti-rheumatism action.Tissue distribution and loranthus avicularin and quercetin belong to the liver and kidney,the traditional understanding is consistent.
