Objective : Through the study to the effect of Anwei Decoction on JAK1/STAT3 signal transduction pathway of CAG rats and its mechanism of action,discussion of the relationship between JAK1/STAT3 signaling pathway and CAG reversal and Anwei Decoction in the treatment of CAG.Methods:82 healthy clean grade male Wistar rats were used,Weighing about 140-170 g or so,10 rats were randomly selected as blank control group(A group).Establishment of MNNG rat CAG model in the remaining72 rats,Respectively as pathological model group(group B),the control group(C group)of the weifuchun,Control group(D group)of the Vitacoenzyme,A small dose group(E group,1 times the dose of adult)of Anwei decoction,The middle dose group(group F,2 times the dose)of Anwei decoction.The high dose group(group G,4 times)of Anwei decoction,At eighth later,2 rats of each model group were randomly selected,put to death,Pathological examination of gastric mucosa,Determine the success of modeling.In the tenth week,the AB two groups were filled with normal saline to fill the stomach,The other groups were given corresponding drugs to fill the stomach,After 4 weeks of drug treatment.Scissor 3 tissue Each rat in theantrum close pylorus to the greater curvature of gastric mucosa,A piece of paraffin section for HE staining,Another two pieces introduction in liquid nitrogen preservation,for Western blotting and real-time PCR test.Results:(1)General situation of rats: the body weight,diet,hair,stool,activity status of the rats in the large,middle dose group and the weifuchun group were significantly better than other groups.(2)perusal for rats:The folds in the model group compared with the other group was flat,rough mucosa,gray color of the surface mucus less,even behold the vascular,there are a few scattered bleeding gastric mucosa were observed.Mucosal improvement in the situation of Anwei decoction soup large,medium dose group is the most obvious.(3)Pathological morphology of gastric mucosa: model group,gastric mucosal atrophy and thinning,the number of natural glands was significantly reduced and arranged in disorder,there are different degrees of inflammatory cell infiltration,mucosal coat thickening.The treatment group and the control group were improved in different degree,especially in the high-dose of Anwei decoction group.(4)western blotting test results:1.JAK1 protein expression: pathologic model group and blank control group comparison expression was significantly increased(P<0.01);compared with the pathologic model group and wefuchun group and Anwei Decoction group decreased the expression and Anwei Decoction high dose group decreased significantly(P<0.01);and comparison with the weifuchun group,Anwei Decoction high dose group protein expression decreased significantly(P <0.01);Anwei Decoction group comparison with the Vitacoenzyme group were decreased significantly(P < 0.01);Anwei decoction high dose group was significantly better than,the small dose group(P<0.01).2.The expression of STAT3 protein: compared with the control group,the expression of protein in model group increased significantly(P<0.01);compared with the model group,weifuchun group and Anweitang group lower,expression of Anwei Decoction high dose group and middle dose group decreased significantly(P<0.01);compared with Weifuchun group.,Anwei decoction high dose group and middle dose group protein expression decreased,there was significant difference(P<0.01),Weifuchun group was lower than low-dose group,but there was no significant difference between them(P>0.05);and vitacoenzyme group,the expression of Anweitang three protein group were significantly reduced,there is significant the difference(P<0.01);compared about Anwei decocotion three group,high dose group and middle dose group was significantly lower than the low-dose group,the difference was statistically significant(P < 0.01).(5)R-T PCR Results: 1.Jak1 gene of Rats detection compared with the blank group compared,the pathologic model group Jak1 mRNA expression increased significantly(P<0.01);compared with the pathologic model group,the treatment group expression decreased significantly(P < 0.01);There was significant difference between the large and middle dose group with the wufuchun group and the Vitacoenzyme group(P<0.05)and Anwei Decoction high dose group is better than the middle dosesoup,small dose group(P<0.05).2.The detection of rat STAT3 gene detection:pathologic model group and blank control group comparison expression was significantly increased(P<0.01);Anwei Decoction group and weifuchun group with reductive to the pathologic model group(P<0.01)and there was significant difference between the Anwei decoction high dose group and the group of the weifuchun(P<0.05);Anwei Decoction high dose group was better than the Vitacoenzyme group(P<0.05);Anwei Decoction in large and middle dose group was superior to low dose group(P<0.05),high dose group and middle dose group had no significant difference compared(P>0.05).Conclusion: 1.Anwei decoction can obviously improve the general state of CAG rats and pathological morphology of gastric mucosa.2.Anwei decoction may regulate apoptosis by inhibiting the JAK1/STAT3 signaling pathway,which can reverse the role of CAG.
