Objective: The experiment is to copy two-kidney-one-clamp renal hypertensive rats'(2K1C-RHR)model and observe the effect of Ziyin Qianyang prescriptions to lower blood Pressure of the hypertensive model rat,and observe the change of protein's expression of AT1R?PKA and CREB in thymus and spleen and content of IL-2 in blood serum.By studying the influence of Ziyin Qianyang prescriptions to the signal path mediated by the hypertensive modal rat's AT1 receptor,to explore effects of nerve,immune and endocrine systems in the pathological of hypertension play an integral role,and the anti-hypertensive mechanism of Ziyin Qianyang prescriptions to the model rat.Further elaborate target of anti-hypertensive mechanism and by Ziyin Qian Yang prescriptions.Methods: To screened 60 successful model rats,rats were divided into model gr oup,western medicine group,traditional Chinese medicine group and Ziyin Qian yang prescriptions high,medium and low group randomly,as well as 10 rats in each of normal control group and sham-operated group,a total of eight groups.To observe testing objective indicators of corresponding treatment in rats in each group For example,protein's expression of AT1R?CREB?PKA in thymus and spleen were measured by the method Immunohistochemical staining,the content of IL-2 in blood serum were measured by the method of ELISA,and observing the pathological changes of the rat's right kidney by HE.Statistical methods: us-ing SPSS18.0 software package for processing,P<0.05 for the difference was stat-i stically significant.Results: 1.The influence of Ziyin Qianyang prescriptions on model rat's systolic blood pressure: after building the model 2W,model group and each treatment group of systolic blood pressure in rats were higher significantly,Compared with control group and normal group,there were statistically significant(P<0.01);and the systolic blood pressure value?160mm Hg,absolute value of difference before and after building ?20mm Hg.2.2K1C-RHR was established successfully: after medication 4 weeks,Ziyin Qianyang prescriptions each dose group(32,16,8 g/kg)?Losartan Potassium Tablets group and Tianma Gouteng group,could significantly reduce systolic blood pressure,compared with model group,there were statistically significant(P<0.01).The influence of Ziyin Qianyang prescriptions on model rat's renal histopathology: Observed from the morphology of kidney,compared with the left kidney,the right kidney of the model group obvious atrophy,surface was uneven,the necrotic areas were yellowish gray and hard non-necrotic areas were dark red;left kidney surface was smooth,dark yellow,volume increased obviously than sham-operation group.HE staining on the pathological displayed,the model group rat glomerulus atrophy,renal tubule degeneration and disordered arrangement;and each group of Ziyin Qianyang prescriptions has different degrees of improvement.Ziyin Qianyang prescriptions groups have certain protective effect to the kidney.The influence of Ziyin Qianyang prescriptions on model rat's serum IL-2:The serum IL-2 of the model group rats were decreased significantly(P<0.01);Compared with model group,serum IL-2 of treated group were significantly higher(P<0.01).3.The influence of Ziyin Qianyang prescriptions to the expression level of AT1 R,PKA and CREB in the thymus,spleen on model rat's: 4.(1)The expression level of AT1R:Expression of spleen and thymus' s AT1 R in the model group rats were increased significantly(P<0.01);compared with model group,the expression of spleen and thymus' s AT1 R in other treated groups were decreased significantly(P<0.01).(2)The expression level of PKA:Expression of spleen and thymus' s PKA in the model group rats were increased significantly(P<0.01);compared with model grou p,the expression of spleen and thymus' s PKA in other treated groups were decr eased significantly(P<0.01).(3)The expression level of CREB:Expression of spleen and thymus' s CREB in the model group rats were increased significantly(P<0.01);compared with model group,the expression of spleen and thymus' s CREB in other treated groups wered ecreased significantly(P<0.01).Conclusion: 1.Ziyin Qianyang prescriptions could anti-hypertension.2.Ziyin Qianyang prescriptions could protect the model rat's kidneys.3.The model rats had immune system disorder,Ziyin Qianyang prescriptions could improve the level of IL-2 inflammatory markers in peripheral blood,combined with its effects on the RAS system;so Ziyin Qianyang prescriptions may played the overall adjustment to the body by through neural immune endocrine system.4.Ziyin Qianyang prescriptions reduced the expression level of AT1 R,PKA and CREB in the thymus,spleen,prompted that influence to lymphocyte AT1 receptor mediated G protein-c AMP-PKA signaling pathway may be one of the body's overall adjustment mechanism of hypertension rats.
