Objectives: The purpose of the study was to observe the effects of XiangLianHuaZhuoJieDu Decoction on ulcerative colitis model rats established by trini-trobenzenesul-fonic acid/ethanol(TNBS/ethanol) through observating the general condition, disease activity index(DAI) scores and the expression of tumor necrosis factor-?(TNF-?)?epidermal growth factor(EGF)try to explore the mechanism of the XiangLianHuaZhuoJieDu Decoction. In order to explore the mechanism for the treatment of UC rats,and provid theoretical and experimental basis in the clinical application.Methods: 60 male Wistar rats were divided into six groups randomly,each group 10 rats. They were marked for the normal group(NG), the model group(MG), the high dose group(HDG), the moderate dose group(MDG), the low dose group(LDG), the SASP group(SASP). All groups except NG were controlled in order to make UC animal models by TNBS/ethanol. 3 days after the building, NG rats in the normal feeding, saline lavage to MG rats, SASP rats using Srlfasalazine Mixed suspension lavage,the Chinese medicine treatment groups used the corresponding drug lavage.After 2 weeks of continuous treatment, observed the state of mind,drinking and diet, activity,weight, excrement characters, and so on and so forth of rats in each group every day.After the treatment, each group of rats were given fasting and water for 24 hours,and recorded their weight.Observed the general view of the colonic mucosa and histopathological changes after all rats were sacrificed.Detected the expression of TNF-? and EGF by PT-PCR and analyse effects of XiangLianHuaZhuoJieDu Decoction on the expression of TNF-? and EGF.Results:1 The general case and DAI scoresCompared with the NG, the general condition of rats became lacking ofenergy, eating less, activity decreased, reaction reduced, weight loss, hair loose and dull, diarrhea or bloody mucus. After the 2 weeks treatment, There were no significant difference between the general condition of MDG and SASP, both lower than the HDG. The HDG rats were close to the NG. LDG were better than MG, and was inferior to MDG and SASP.The DAI scores : In terms of disease activity index score, there was statistically significant difference(P<0.05) between the treatment group HDG(0.37±0.35),MDG(0.90±0.32),LDG(1.50±0.62),SASP(0.63±0.35)and MG(2.50±0.47). There was statistically significant difference(P<0.05)between HDG and MDG,LDG. There was no statistically significant difference(P>0.05) between SASP and HDG, MDG. There was statistically significant difference(P<0.05) between LDG and SASP, MDG.2 The general observation and the histopathologicalThe MG was the worst. The HDG was the best which was closely to the NG. The SASP were better than MDG and LDG while worse than HDG. LDG were better than MG, but was inferior to MDG.Histopathological observation: Mucosa in NG was smooth and lustrous.Mucosal glands in MG,which were infiltrated by a large number of inflammatory cells, along with mucosal hyperemia and edema,were damaged.Mucosal destruction situation in LDG was better than MG. There were basically complete mucomembranous structure in HDG,which was closed to NG.The situation of SASP was inferior to HDG.The outcome of MDG was better than LDG,but inferior to HDG and SASP.3 The expression of TNF-? and EGF of ratsCompared with the NG(0.21 ± 0.02), the expression of TNF-? in MG(0.55 ± 0.03)were significantly increased, and there was statistically significant difference between them(P<0.05). Compared with the MG, the expression of TNF-? in each treatment group were decreased obviously, there was statistically significant difference between them(P<0.05); The HDG(0.31± 0.03) compared with the MDG(0.40 ± 0.02), LDG(0.48 ± 0.02)and SASP(0.36 ± 0.04), Significant difference can be observed(P<0.05); TheSASP compared with the MDG and LDG, there was statistically significant difference(P<0.05); MDG higher than that of LDG, the difference was statistically significant(P<0.05).Compared with the NG(0.69±0.04),the expression of EGF in MG(0.24±0.04)were significantly decreased, and there was statistically significant difference between them(P<0.05). Compared with the MG, the expression of EGF in each treatment group were decreased obviously, there was statistically significant difference between them(P<0.05);The HDG(0.56 ± 0.04)compared with the MDG(0.42 ± 0.04)and LDG(0.33 ± 0.03), Significant difference can be observed(P<0.05); The HDG compared with the SASP(0.55±0.03), no statistically significant difference( P>0.05); The SASP compared with the MDG and LDG, there was statistically significant difference(P<0.05);LDG was lower than MDG wit statistically significant difference(P<0.05).Conclusion:1 XiangLianHuaZhuoJieDu Decoction can improve the general situation of UC rats, also have effect on the alleviation of pathological damages in rats model, and promote treatment of the ulcer. The efficacy showed a certain amount-effect relationship between efficacy and medicine dose.2 The mechanism of action of XiangLianHuaZhuoJieDu Decoction to treat UC is probably likely to improve expression of EGF and decrease expression of TNF-?, inhibit the infiltration of inflammatory cell effectively,repair the damage of intestinal mucosa, so as to achieve the purpose of treatment of UC.
