First, with the rapid development of our society and economy, people's living standard has been improved significantly, increasing obesity, so it is increasingly important to study and propose effective intervention and prevention programs. According to preliminary study about the relationship between human hunger and feeding, people have eaten the prebiotic which can not be absorbed in human's intestinal but can be metabolized by commensal bacteria, the feeling of hunger was became weakened or even disappear, on this research foundation we proposed a scientific hypothesis about " the feeling of hunger from flora " and flexible fasting technology program.In our study, we focus on physiological and biochemical indices and subjective of many volunteers, to evaluate whether this flexible technology as a safe and effective methods for weight loss. During the seven-day study Flexible fasting, volunteers used a flexible refraining solid beverages replaced a meal, and drinking, fruit or vegetables by the needs of everyone,we also checked the blood biochemical parameters, weight blood pressure, and heart rate were recording of volunteersdaily physical condition.Our results indicated that after seven days flexible fasting all volunteers in the flexible testing process, body weight, high-quality sleep, was became an ideal changing, heart rate, blood and indicators of liver and kidney function pressure was not significant changing, all the whole in good spirit.These results suggest that Flexible fasting is a good method of a physiological controlling weight loss by refraining intestinal flora, and it could effectively controls weight, reduces obesity and many chronic diseases, provides a valuable reference scheme by this method. Currently, there are many outpatients was specialized on controlling weight in hospitals using flexible fasting. All this results reflect the success of translational medicine research as an important application in clinical practice.Second, molecular hydrogen is a minimum in recent years, studies have found that hydrogen molecules with a clear physiological effects, can improve the intestinal microenvironment, in theory, can enhance the effect of the flexible bigu. At present, for hydrogen research focused on its efficacy but less in the method of application. In this study, we focus on hydrogen and hydrogen production kinetics rods volatile hydrogen gas in a systematic study, it is important to the development of the hydrogen molecular medicine and clinical application of health.In the experiment, first, seven unopened hydrogen rods were placed in a plastic bottle filled with distilled water, soaked for 6 hours to measure the hydrogen content, repeat steps 10 times to determine their degree of stability about hydrogen and losses. The results showed that the amount of the first time hydrogen production mean(0.75 ± 0.11ppm) is much higher than the next nine hydrogen production, and hydrogen production(0.24 ± 0.02ppm), the other nine times( 0.22 ± 0.03ppm) only a difference of 0.02 ppm. Thus, the stability and the degree of loss of hydrogen rods could meet the requirements of the experiment.Next, hydrogen rod kinetics analysis of hydrogen production, hydrogen to 5 bar into 5 bottles filled with distilled water in plastic bottle, were placed in a water bath pot 20, 40, 60? and were 2, 4, 6, 8, 10 hours of soaking hydrogen, and hydrogen content were measured oxidation reduction potential at various points in time, the results show that at a certain temperature, the hydrogen content of the hydrogen-rich water rises with increasing time oxidation-reduction potential over time increases.After analyzing the dynamics of hydrogen stick hydrogen production, we focus on hydrogen volatile dynamics analysis. The prepared hydrogen-rich water divided into two groups, there is no bar group as control and bar group, the group does not stick after opening the hydrogen stick out, stick-free group in the bar after opening the hydrogen removed, 0min after opening the lid, 10 min, 30 min, 1h, 2h, 5h, 12 h, 24 h, 30 h, 48 h, 72 h were measured hydrogen content, two group not closing lid and kept at 25?constant temperature water bath. The results found that hydrogen-rich water rod group in the fall to around 0.50 ppm began keeping a constant hydrogen content without rod group at 72 hours, and the amount of hydrogen has been reduced to 0ppm.Then use the zebrafish to do experiment about the acute inflammatory antioxidant effects of paraquat poisoning by using hydrogen-rich water. The result showed zebrafish lived in hydrogen-rich water the lifetime became shorter than in control group, analyzing the possible reasons was the hydrogen-rich oxygen content in water is low. By detecting three different hydrogen-rich water and control water or raw water of oxygen found that in water oxygen hydrogen-rich water or raw water is lower than the control group(P <0.01, P <0.01, P <0.01). Through hydrogen stick to study the kinetics of hydrogen-rich oxygen content in water by the method same as hydrogen production with hydrogen rod kinetic. The results showed that under certain circumstances the temperature, oxygen content increases over time while reducing overall fooled.Third, the intestinal flora disorders such as obesity or chronic diseases,, the body's inflammatory response induced by endotoxin into the blood, prompting upregulate the expression of antimicrobial peptides Hepcidin, while having another identity Hepcidin: iron regulatory protein, which is involved in the body inflammation, also involved in the body's iron metabolism, there may be intrinsic link between the two terms, the intersection point of contact is likely to focus on hypoxia and inflammation.This study was to investigate the different ways movement for Hepcidin expression, and comparative analysis with closely related indicators of iron metabolism, hypoxia response trend factors, inflammatory cytokines and antimicrobial activity related indicators. Hepcidin may further confirmed the intrinsic link itself with the dual biological activity.Results indicated hypoxia-induced failure of the body's inflammatory intense response while body in 4000 m altitude, but in 8000 m altitude and severe hypoxia upregulated by HIF-1? levels, stimulate and increase Hepcidin expression, they have the full iron negative regulator of metabolic effects, so that iron metabolism have a serious disorder, resulting in anemia, at the same time accompanied by inflammation significantly intensified.In summary, the present study for the obesity problem currently, in the light of the early research in our laboratory inspired new ideas for the origin of obesity, in collaboration with some hospitals, we confirm that the method flexible refraining could be applied in physiological weight loss and provided an effective solution to the problem of obesity. On this basis, in view of domestic and international study found that obesity and inflammation caused by intestinal flora disturbance is closely relation to the human body, so, while we study on hydrogen-rich water that has a good antioxidant and anti-inflammatory effection, we also study on hydrogen-rich water generating hydrogen and hydrogen-volatile kinetic characteristics, in order to further optimize the hydrogen-rich water for health improvement provide a reference, we also study on inflammation which closely related to hepcidin hepcidin expression and body movement characteristics under hypoxic conditions. the results made an important foundation for controlling inflammation reaction and be good for improving the body's health.
