Objective:Explore the relationship between VaD and vitD.Observe the effect of vit D deficiency and vit D expression in rat model of SYN on the expression of SYN and GAP-43,to investigate the effect of vit D on the learning and memory ability of rats and its effect on synaptic plasticity.Methods: 1.Clinical research:a parallel,randomized and controlled experimental scheme,to choose between the traditional Chinese Medicine Hospital of Liuzhou city from December 2014 to December 2015,60 cases of patients with 30 cases of VaD patients and the diagnostic standard of health examination center of normal serum,compared two groups of 25(OH)D3.2.According to the methods of Qinghua lee,The 50 SD rats were divided into(the control group,n=10)?(the model group,n=10),(Low dosed group,n=10),(Middle dosed group,n=10),(High dosed group,n=10)for Vit D deficiency feed 9 weeks,light adopts ultraviolet(uv)light bulb,circadian rhythms 12hours/12 hours.Vit D supplements to fill the stomach in the 10 weeks after 2weeks using Morris water maze method is used to test the learning and memory ability of rats,and within the prescribed time period to be put to death in the rat brain tissue.Enzyme-linked immunosorbent(ELISA)method to detect the contents of 1,25 hydroxy Vit D3;Using Western blot detection of SNY and GAP-43 protein expression.Data from the SPSS23.0 statistical analysis software,the data using mean±standard deviation,said the comparison between the two sample mean using t test,compared with single factor analysis of variance between groups(one-way ANOVA).Variance together when usingLSD method and variance is to use Dunnett 's T3 law to P<0.05 for the difference was statistically significant.Results:1.Rats with the increase of training days,average escape latency period shorten,rats search hidden platform edge type,ondemond strategy from the start,gradually into line and trend model.Model group escape latency in training day compared with the escape latency of blank control group,with statistical significance(p?0.05).Blank control group and Vit D target quadrant 1 time through(the third quadrant)there were significant differences between(p ?0.05).Target quadrant(the third quadrant)across,in terms of a lack of Vit D group(1.90±1.37)significantly lower than the blank control group(5.10±1.91),comparing the two groups was statistically significant(p?0.05).2.Compared with blank control group,Model group of SNY protein expression increased(P<0.05);Compared with Model group,high dose group of SNY protein expression were higher(P<0.05);Compared with blank control group,Model group GAP-43 protein expression increased(P<0.05);Compared with Model group,high dose group of GAP-43 protein expression were higher(P<0.05).Conclusion:1.Vit D deficiency may be related to the ability of learning and memory impairment in rats.2.Vit D supplements for a long period of time will benefit the reshaping of the synapses,can improve Vit D deficiency rats learning and memory ability.3.Vit D is the role of the ability to improve learning and memory of rats,may be associated with increased synaptic protein portfolio and GAP-43 protein expression.Day 2 high dose group of escape latency(35.65±32.09 s)with Model group escape latency(55.28 ± 38.07 s)compared with statistical significance(p?0.05).High dose group and a lack of Vit D in the target quadrant only(the third quadrant)through the number of significant difference(p > 0.05).
