Objective: To compare the pregnancy outcomes in preeclampsia patients and normotensive pregnancy women,and to investigate the expression and clinical significance of dysferlin and affixin in peripheral blood and placenta and to observe the difference of both,making dysferlin and affixin become biomarkers,as a prediction of preeclampsia,in the purpose of providing reliable objective basis for early diagnosis.Methods:1 The study subject: According to the diagnostic criteria of preeclampsia,randomly selecting patients with preeclampsia from the second hospital of Hebei medical university from 2014-12 to 2015-12.In the 71 cases,38 pregnant women suffer from early onset preeclampsia,33 pregnant women suffer from late onset preeclampsia,and 33 normal pregnant women were selected randomly as the control.2 Sample collecting: After admission,3ml of fasting venous blood samples(separated plasma by 2000 rpm and 10 minutes)was collected before treatment,was stored at-20? refrigerator for assaying the concentrations of plasma dysferlin and affixin.Immediately after delivery of the placenta should charge maternal surface of placental tissue and recent the umbilical cord root,about 1cm3,after saline flush with absorbed by filter paper,at room temperature for 48 h in 10% formalin and conventional dehydrated paraffin embedded.3 The experiment indicators: All of the subjects were detected the expression of dysferlin and affixin by enzyme-linked immuno sorbent assay(ELISA)and immunohistochemistry.We recorded age,gestational week at delivery,height,weight,blood pressure and other indicators.4 Statistical methods: The results were analyzed by the SPSS 21.0 software.Results:1 The birth weight in control group was(2740.76±457.00)g,it in early onset group was(1363.03±453.29)g,and it in late onset group was(2541.97±680.78)g.The induction of labor rates of groups were 0,32%,0,respectively.The premature birth rates of groups were 6%,100%,39%,respectively.The low birth weight rates of groups were 6%,100%,45%,respectively.The birth weight was decreased in women with preeclampsia compared to women with normal pregnancy(P<0.05).And it in the early onset preeclampsia was lower than it in late onset preeclampsia(P<0.05).The induction of labor rate,premature birth rate,low birth weight rate were increased in women with preeclampsia compared to women with normal pregnancy(P<0.05).And it in the early onset preeclampsia was higher than it in late onset preeclampsia(P<0.05);2 Plasma level of dysferlin in normotensive and preeclampsia were undetected with ELISA.Plasma level of affixin in normotensive was(4.13±0.58)ng/ml,it in early onset preeclampsia was(3.07±0.62)ng/ml,it in late onset preeclampsia was(3.65±0.74)ng/ml,Plasma levels of affixin were decreased in women with preeclampsia compared to women with normal pregnancy(P<0.05).And it in the early onset preeclampsia was lower than it in late onset preeclampsia(P<0.05);3 The expression level of dysferlin in normotensive was(7.49±0.83),it in early onset group was(4.29±0.95),it in late onset group was(5.53±1.31),the levels of dysferlin were decreased in women with preeclampsia compared to women with normal pregnancy(P<0.05).And it in the early onset preeclampsia was higher than it in late onset preeclampsia(P<0.05).The expression level of affixin in control group was(7.44±0.91),it in early onset group was(4.88±0.49),it in late onset group was(4.71±0.49),the levels of affixin were decreased in women with preeclampsia compared to women with normal pregnancy(P<0.05);4 The expression level of dysferlin and affixin in placental syncytiotrophoblast were correlated positively,and the correlation coefficient r=0.67(P<0.01).Conclusion:1 The birth weight of preeclampsia group was lower than the control group and it in the early onset preeclampsia was lower than it in late onset preeclampsia.The premature birth rate,induction of labor rate and low birth weight rate of preeclampsia group were higher than the control group and it in the early onset preeclampsia was higher than it in late onset preeclampsia.So preeclampsia contributes to poor outcome,especially early onset preeclampsia.2 Plasma levels of dysferlin were not detected of both groups.Plasma levels of affixin were decreased in women with preeclampsia compared to women with normal pregnancy,which shows that the level of dysferlin can not be used as a biological marker,while affixin in plasma of venous blood can be used as a biological marker,as a prediction of preeclampsia.3 Dysferlin was mainly expressed in cytomembrane of syncytiotrophoblast in both groups.The levels of dysferlin were decreased in women with preeclampsia compared to women with normal pregnancy;especially it in the early onset preeclampsia was lower than it in late onset preeclampsia.Affixin was mainly expressed in cytomembrane of placental syncytiotrophoblast in both groups,and the expression of affixin in placenta of patients with preeclampsia complicating pregnancy was increased as the disease worsened.All above show dysferlin and affixin play a role in preeclampsia.4 The expression levels of dysferlin and affixin in syncytiotrophoblast were correlated positively,and the interaction effects between them may relate to the development and progression of preeclampsia complicating pregnancy.
