Objective: To investigate the expression and the correlation of Shh and Gli-1 which were the initial factor and nuclear transcription factor in SHH signaling patyway among different levels' neuroastrocytoma and normal brain tissue.Methods:1 To return the neuroastrocytoma and brain contusion tissue from May 2014 to May 2015 which were all from the Neurosurgery of the Fourth Hospital of Hebei Medical University.The neuroastrocytoma tissue 50 cases(36 cases high grade glioma tissue and 14 cases low grade glioma)and the normal brain tissue 10 cases were all fixed by formaldehyde.The neuroastrocytoma tissue 20 cases(10 cases high grade glioma tissue and 10 cases low grade glioma)and the normal brain tissue 8 cases were all frozen in-80? refrigerator(the normal brain tissue was all from craniocerebral injury patients;before operation,we had told the use of the tissue to patients' relation and got their agreement).2 According to the central nervous system tumor classification criteria of WHO,high grade glioma was the ?~?grade and low grade glioma was the ?~?grade.3 Using the Immunohistochemistry to detective the different expression of Shh and Gli-1 on protein level.4 Using the RT-PCR to detective the different expression of Shh and Gli-1 on gene levels.5 Using the statistical software SPSS 21.0 to analyze data.Results: 1 Immunohistochemistry results: Shh positive sign located in cell membrane?cell cytoplasm.Gli-1 positive sign located in cell cytoplasm?cell nucleus.In high grade glioma,the positive rate of Shh was 61.1%(22/36);In low grade glioma,the positive rate of Shh was 57.1%(8/14);In normal brain tissue,the positive rate of Shh was 10%(1/10).These showed significant growth(P<0.05).In high grade glioma,the positive rate of Gli-1 was 69.4%(25/36);In low grade glioma,the positive rate of Gli-1 was 28.6%(4/14);In normal brain tissue,the positive rate of Gli-1 was 10%(1/10).These also showed significant growth(P<0.05).Among them,the two contrast,between high grade and low grade glioma,the positive rate of Shh showed insignificant growth(P>0.0125);between high grade glioma and normal brain tissue?low grade glioma and normal brain tissue,the positive rate of Shh showed significant growth(P<0.0125).Between high grade and low grade glioma?high grade glioma and normal brain tissue,the positive rate of Gli-1 showed significant growth(P<0.0125);between low grade glioma and normal brain tissue,the positive rate of Gli-1 showed insignificant growth(P>0.0125).2 RT-PCR results: In high grade glioma,the relative expression of Shh was 0.126±0.020;In low grade glioma,the relative expression of Shh was 0.113±0.045;In normal brain tissue,the relative expression of Shh was 0.061±0.039.These showed significant growth(P<0.05).In high grade glioma,the relative expression of Gli-1 was 0.141±0.032;In low grade glioma,the relative expression of Gli-1 was 0.074±0.044;In normal brain tissue,the relative expression of Gli-1 was 0.040±0.024.These also showed significant growth(P<0.05).Among them,the two contrast,between high grade and low grade glioma,the relative expression of Shh showed insignificant growth(P>0.0125);between high grade glioma and normal brain tissue?low grade glioma and normal brain tissue,the relative expression of Shh showed significant growth(P<0.0125).Between high grade and low grade glioma?high grade glioma and normal brain tissue,the relative expression of Gli-1 showed significant growth(P<0.0125);between low grade glioma and normal brain tissue,the relative expression of Gli-1 showed insignificant growth(P>0.0125).3 The correlation analysis: In immunohistochemistry,the correlation factor was 0.926(P<0.05);In PCR,the correlation factor was 0.588(P<0.05).Conclusions:1 The expression of Shh and Gli-1 in glioma tissue were obviously higher than these in normal tissue.This prompted that the SHH signaling patyway was generally actived in glioma.2 In immunohistochemistry and RT-PCR,the linear correlation was showed between Shh and Gli-1's expression.This prompted that both Shh and Gli-1 participated in glioma's occurrence and development.3 In immunohistochemistry and RT-PCR,the expression of Shh showed insignificant growth between different grade glioma,the expression of Gli-1 showed significant growth between high grade and low grade glioma,however.These prompted that a lot of signal protein which was belong to SHH signaling pathway might be mutation in high grade glioma,such Ptch?Smo and so on.
