Objective:By observing the level of rerum ET-1?BNP in rats with Myocardial Infarction and the intervention by Xin Tong Tai granular formulation, and to study the targets and inhibition mechanism of ventric remodeling by Xin Tong Tai granular formulation.Methods:100 SD Rat model of acute myocardial ischemia was established by ligating the left anterior descending branch of coronary artery.Then they were randomly divided into six gronps:model group, control group, Xintongtai low-dose group, Xintongtai middle-dose group, Xintongtai high-dose group, Shexiangbaoxinwan group.From the second day after establishing the model, corresponding drugs were given to each treatment group through administration by gavage, The same volume of distilled water were given to the model group and the control group. It was once a day, last for two weeks.Hematoxylin-eosin staining was used to observe the change of myocardial morphology under light microscope, TTC staining method was used to detecte the myocardial infarct area. Serum endothelin 1(ET-1)?Brain natriuretic peptide(BNP) tested through ELISA Kit.Results:1.The changes in myocardial tissue morphological:Control group:the arrangement myocardial cell was regular, some cells were mild edema; Nuclear was rod; stripes were visiable; Intercellular substance was well-distributed; Model Group:The arrangement of myocardial cells were disordered, The cells were edema; stripes got fractured; mesenchyme was broadened; vasculars were congestive, more inflammatory cells were infiltrated; large numbers of fibrosis were formed in the infarction area; Xintongtai low-dose group:The myocardial cells arranged irregular, but compared with model group:slightly neat, the lower myocardial cells were edema, mesenchyme was broadened; fewer inflammatory cells were infiltrated, fibrosis were visible in part of the myocardial infarction area? Xintongtai middle-dose group:compared with model group, the arrangement of myocardial cells were regular, stripes got fractured occasionally, part of myocardial cells were edema, moderate inflammatory cells were infiltrated; a litter of fibrosis were visible in myocardial infarction area; Xintogtai high-dose group:The arrangement of myocardial cells were more order, mild edema, a small amount of inflammatory cells were infiltrated; there was a small amount of inflammatory cells were infiltrated; there was a small amount of fibrosis in myocardial infarction area; Shexiangbaoxinwan group:The arrangement of myocardial cells were more regular, some cells were edema, stripes got fracture partly; a small amount of inflammatory cells were infiltrated; a small amount of fibrosis were visible in infarction area; 2.Myocardial infarction area:Infarction area was not seen in control group; Compared with model group, The area of treatment groups were decreased, among them, the difference was significant when compared with the Xintongtai low-dose group, but it was obvious different when compared with the Xintongtai middle-dose group, Xintongtai high-dose group and Shexiangbaoxinwan group; Compared with Xintongtai low-dose group, The infarction area in Xintongtai middle-dose group, Xintongtai high-dose group were silightly reduced, but there was no difference among them(P>0.05), however, the Shexiangbaoxinwan group was statistically significant(P<0.05),but there was no difference among Xintongtai low-dose group, Xintongtai middle-dose group, Xintongtai high-dose group(P>0.05).3.IVT and IVST:compared with control group, the thickness of the model group and the treatment groups have increased, and the difference was statistically significant(P<0.01); compared with model group, the thickness of the treatment groups have reduced, and the difference was statistically significant(P<0.01); compared with Xintongtai low-dose group, Xintongtai middle-group> Xintongtai high-dose group and Shexiangbaoxinwan group, there was statistically significant difference among those groups, the difference in this index among Xintongtai middle-dose group.. Xintongtai high-dose group and Shexinagbaoxinwan group was not significant(P>0.05);4.The level of ET-lin serum:compared with control group, the amount of serum ET-1 was increased siginificantly in treatment group and model group(P<0.01), compared with model group, the amount of this index was reduced in treatment group(P<0.01), compared with Xintongtai low-dose group, the amount of this index was reduced in Xintongtai middle-dose group?Xintongtai high-dose group and Shexinagbaoxinwan group(P<0.01), the difference in this index among Xintongtai middle-dose group? Xintongtai high-dose group and Shexinagbaoxinwan group was not significant(P>0.05);5.The level of BNP in serum:compared with control group, the amount of serum BNP was increased siginificantly in treatment group and model group(P<0.01), compared with model group, the amount of this index was reduced in treatment group(P<0.01), compared with Xintongtai low-dose group, the amount of this index was reduced in Xintongtai middle-dose group, Xintongtai high-dose group and Shexinagbaoxinwan group(P<0.01), the difference in this index among Xintongtai middle-dose group? Xintongtai high-dose group and Shexinagbaoxinwan group was not significant(P>0.05).Conclusion:Xintongtai granular formulation can protect the ischemic myocardium, reduce the infiltrated inflammatory cells, inhibit the fibrosis of mesenchyme, reduce Myocardial infarction area?IVT and IVST, suggest that it maybe by lowering the levers of serum ET-1 and BNP to suppress the RAAS system, then implement the inhibitation of ventric remodeling.
