Whether it is early or late in tumor, physicians, cancer patients and their families as well as the whole society pay special attention to the problem in looking for a substance to auxiliary treatment of chemotherapy drugs to reduce side effects of chemotherapy drugs.Confirmed by a large number of data and experimental data on the related barley seedling powder abroad and at home, it is found that barley seedling powder has the effects on improving immunity, antioxidant, anti-aging, anti-tumor and reducing blood sugar. There is a kind of medicinal materials as pharmaceutical as well as food, which is called "the homology of medicine and food", such as Chinese wolfberry, coix seed, honeysuckle, lotus leaf, mulberry leaf, etc. A large number of studies showed that the homology of medicine and food also has the effects on improving immunity, anti inflammation, antioxidant, anti-aging, anti-tumor and so on.Materials used in this experiment-Composite Barley Seedling Powder including two kinds of products: Wheat Green Power and Wheat Adlay Power. Wheat Green Power is an product of auxiliary function during the cancer chemotherapy and Wheat Adlay Power is an products of auxiliary function for advanced cancer patients. It is expected to solve the major medical problems, which is of great significance to the study of the adjuvant treatment of cancer.This experiment includes two parts:Part one: The auxiliary function of Wheat Green Power on chemotherapy in tumor bearing mice and its mechanism. Main components of Wheat Green Power are Wheat and Green Tea, Honeysuckle, Mulberry leaf, Lotus leaf and other plant ultrafine powder composition.1. Composition determination: The experiments using the national standard method in the determination of main nutritional components and active ingredient content of Wheat Green Power. Measurement results show that the contents(g/100g)of protein,fat,total,cellulose and carbohydrate in Wheat Green Power were 12.33,6.75,37.40,25.70. The contents of polysaccharides, flavonoids and chlorophyll(mg/g) in Wheat Green Power were 21.54,8.17,19.79.2. In vitro experiment: MTT method was used to determine the inhibitory effect of different concentrations of Wheat Green Power(0.05, 0.10, 0.15, 0.20g/ml) on S-180 tumor cells and mouse peritoneal macrophage proliferation and the suitable dosage for vivo experiment was selected. Measurement results show that Wheat Green Power has certain inhibitory effect(Inhibition rate in 37.3%-72.9%) on S-180 tumor cells in vitro and proliferation effect(Proliferation rate in 93.40%-98.82%),there was no difference between the four concentrations of wheat green powder and the immune enhancing agent(Proliferation rate is 90.33%).The suitable dosage for vivo experiment was 0.125 g/ml.3. In vivo experiment: In vivo experiments were carried out by using the suitable dosage, S-180 sarcoma models of mice was built up. Chemotherapy drugs(5-FU) was used for 3 days in a row while Wheat Green Powder for 15 days continuously. Theinfluence of the quality of tumor, the liver spleen index and blood cells in tumor bearing mice was observed afterwards. Measurement results show that chemotherapy with Wheat Green Power can significantly improve the tumor inhibition rate was 74.3%, the chemotherapy group was 45.9%, the general state of mice is better than chemotherapy group, and significantly increased the spleen index and the number of mouse neutrophils, monocytes, lymphocytes and eosinophils(P < 0.05). From the analysis of the immune organs and immune cells, visible Wheat Green Power could effectively reduce 5-FU induced lymphocyte, monocyte and granulocyte counts, so as to alleviate the 5-FU induced immune cells decreased; For another, Wheat Green Power can significantly improve the eosinophil absolute value, while significantly reducing the basophils absolute, that Wheat Green Power has certain anti-inflammatory response.4. Conclusion: Wheat Green Power has certain inhibitory effect on S-180 sarcoma cells In vitro and in vivo and it can reduce the toxic and side effects caused by chemotherapy drugs. Wheat Green Power has certain anti-inflammatory response.Part two: The auxiliary function of Wheat Adlay Powder on late stage tumor bearing mice and its mechanism. Main components of Wheat Adlay Powder are wheat and Lily, coix seed, polygonatum, fungus and other plant ultrafine powder composition.1. Composition determination: The experiments using the national standard method in the determination of main nutritional components and active ingredient content of Wheat Adlay Powder. Measurement results show that the contents(g/100g)of protein,fat,total,cellulose and carbohydrate in Wheat Adlay Powder were 10.50,5.17,34.99,26.22. The contents of polysaccharides, flavonoids and chlorophyll(mg/g) in Wheat Adlay Power were 24.89,5.30,12.19.2. In vitro experiment: MTT method was used to determine the inhibitory effect of different concentrations of Wheat Green Power(0.05, 0.10, 0.15, 0.20g/ml) on S-180 tumor cells and mouse peritoneal macrophage proliferation and the suitable dosage for vivo experiment was selected.Measurement results show that the inhibition rate of Wheat Adlay Power on S-180 tumor cell was 48.77%-87.60% and the effect of Wheat Adlay Power on proliferation macrophages(the proliferation rate was 23.94%-41.49%) was situated between 5-FU(the proliferation rate was 11.01%)and levamisole hydrochloride(the proliferation rate was 86.17%). The suitable dosage for vivo experiment was 0.15 g/ml.3. In vivo experiment: In vivo experiments were carried out by using the suitable dosage and Mouse S-180 late stage tumor model was built up, intragastric administration of Wheat Adlay powder(0.15g/ml)for 10 days continuously in vivo. By observing the influence of Wheat Adlay powder on the quality of tumor in tumor bearing mice, the quality of the liver spleen, blood cells, calculating the inhibition rate and the index of liver and spleen. Measurement results show that it was found that Wheat Adlay powder can inhibit the tumor growth in vivo of the late stage mice of tumor, its inhibitory rate reaches 81.1%; Compared with tumor control group, it Can significantly improve the number of mononuclear cells, eosinophils(P < 0.05), and reduce the number of basophils(P < 0.05), compared with the experimental group's, it have not diverged very much before and after the experiment. The experimental results show that Wheat Adlay powder may be through a certain anti hypersensitivity reaction to improve advanced cancer of inflammatory cytokines increased in cachexia reaction.4. Conclusion: Wheat Adlay Power has certain inhibitory effect on S-180 sarcoma cells in vitro and in vivo and it can balance the function of immunity, aslo make inflammatory reaction relieved to a certain extent, and improved the cachexia status on late stage cancer. |
