Objective: We found that the incidence of depression and anxiety in patients with type 2 diabetes were significantly increased,and insulin resistance,making them difficult to control the blood glucose.The TCM believes that the liver adjustable smooth emotions.On the basis of clinical guidance,mentor based from the theory of liver treatment of diabetes syndrome differentiation model,advocated the using of Sini san to reconcile with emotion and regulation in patients with hormone balance and to control blood sugar.We intend to study the effect of four inverse scattered treatment on glucocorticoid and liver 11 beta-HSD1 and GR protein expression in rats with Sini san for the treatment of type 2 diabetes mellitus with insulin resistance.Methods: Establishment of type 2 diabetic rat model by high glucose and high fat diet combined with intraperitoneal injection of stretozocin.The body weight,fasting blood glucose,blood glucose,serum insulin,plasma corticosterone and insulin sensitivity index were detected,and Blot Western method was used to detect the 11 beta-HSD1 and GR protein in the liver to observe the therapeutic effect of the Sini san on experimental diabetic rats.Results: After administration,Sini san can control the increase of body weight of diabetic rats(P<0.05);have reduced the role of fasting and random blood glucose(P < 0.01),and high dose of Chinese medicine group hypoglycemic effect is better;reduced serum insulin level and insulin resistance index(P<0.01);reduce the plasma concentration of corticosterone.In model group(p < 0.05),the liver of the 11beta-HSD1,GR proteinexpression was up-regulated(p<0.01),the treatment of traditional Chinese medicine the expression of the two proteins were down that Sini san may through modulation of signaling pathways(p < 0.01),thereby affecting protein molecular level so as to improve insulin resistance,lower blood sugar,plays a role in the treatment.Conclusion(s): Sini san can reduce the content of diabetic rat glucocorticoid and decreased in the liver of the 11beta-HSD1 and GR protein expression;Sini san can also control the increase of body weight of rats,reduce blood glucose,insulin and insulin resistance index.It is suggested that the Sini san may be through the link between the effects of 11 beta-HSD1--GCs--GR,up to suppress hepatic glucose production,lowering blood glucose,improve insulin resistance in the role.
