Objective:The main effective components of The Mahuang Cook Pulvis as the index,to determine the optimal conditions of The Mahuang Cook Pulvis.Determine the best dosage for clinical use.The Mahuang Decoction and The Mahuang Cook Pulvis by animal pharmacodynamics comparative study,To verify whether The Mahuang Cook Pulvis and The Mahuang Decoction efficacy consistent.To Provide scientific basis for clinical application.Methods:(1)Refer to The Mahuang Cook Pulvis of related materials,formulation research,to determine the main effective components.(2)To establish a method for determination of The Mahuang Decoction main effective components.And to determine the content of The Mahuang Decoction.(3)The content of The Mahuang Decoction of the main active components as the assessment index,use the Single factor method and orthogonal test multiple factor method,The Mahuang Cook Pulvis preparation,decocting process optimization research.(4)The Mahuang Decoction and The Mahuang Cook Pulvis pharmacodynamics comparative study:(1)The observation of The Mahuang Decoction and The Mahuang Cook Pulvis affected by dry yeast suspension induced fever model,compare the antipyretic effect.(2)Using sequential method and The Mahuang Cook Pulvis on the inhibitory effect of mouse model of cough induced by ammonia stimulation.(3)To observe the decoction and Cook Pulvis on by xylene induced inflammation of mouse auricle swelling model and carrageenan glue induced mouse paw edema model effects and anti-inflammatory effect.Results:(1)Prescription research results: to determine ephedrine hydrochloride,pseudoephedrine hydrochloride,amygdalin as main effective components of The Mahuang Cook Pulvis.(2)Established the HPLC method for the determination of The Mahuang Decoction method of the main active component of ephedrine hydrochloride,pseudoephedrine hydrochloride,amygdalin content powder: with the mobile phase of methanol and 0.1% phosphoric acid for 0min(6:94-50min(10:90)-75min(10:90)-75 min 6:90;at a flow rate of 1ml/min;detection wavelength was 215 nm,column temperature of 25°C;sample size: 20?L;after running 10 minutes.The number of theoretical plates,according to the calculation of ephedrine hydrochloride of not less than 3000,according to the calculation of amygdalin,not less than 7000.(3)To determine The Mahuang Cook Pulvis the best preparation process: crushing 50 mesh sieve,a non-woven bag.Determine The Mahuang Cook Pulvis for boiling the best decocting process: take equivalent of The Mahuang Decoction of herbs pieces of half dose of The Mahuang Cook Pulvis into no woven bags,15 times the amount of water,boiling 9min,fried provided 1,filtering,concentrating to 50 ml.determine the recommended use of the powder for the treatment of half of the dose,that is 12 g.(4)Decoction and pharmacodynamics of The Mahuang Decoction :Antipyretic experiment:(1)The Mahuang Cook Pulvis high group,The Mahuang Cook Pulvis in dose group,The Mahuang Cook Pulvis low-dose group and decoction group antipyretic effect difference between the two groups was not statistically significant(P>0.05),that traditional decoction and Cook Pulvis effect had no significant difference.Compared with the blank group,there were significant differences(p<0.01).That Cook Pulvis and Decoction is significant antipyretic effect.(2)Cough experiment:The Mahuang Cook Pulvis high dose group R values for these different,there is a significant effect in relieving cough;The Mahuang Cook Pulvis dose of R value is 14.45%,has the effects of relieving cough;The Mahuang Cook Pulvis low-dose group R was 123.1%,antitussive effect is not obvious.The traditional decoction group R was 138%,have a antitussive effect.(3)The anti-inflammatory experiments:(1)cooked powder in dose group,cooked powder low-dose group and decoction group mice auricle swelling inhibition rate,the difference was not statistically significant(P>0.05),There was significant difference between the high dose group and the traditional decoction group(P<0.05),and there was significant difference between the model group and the model group(P<0.01).Boiling powder in dose group,cooked powder low-dose group and decoction group of mouse foot swelling inhibition rate,the difference was not statistically significant(P>0.05),compared with model group,there was significant difference(P<0.01).Conclusion: Accurate determination method established in this study Mahuangtang main effective component content is simple,have a good reproducibility.The Mahuang Cook Pulvis has a simple preparation process,good operability.Determine the clinical recommended.preliminaryanimal pharmacodynamic comparison results show,the Cook PulvisandDecoction in relieving cough,antipyretic,anti-inflammatory effects no significant difference.The medicinal dosage only half or less of the traditional decoction.will be used to promote the spread of the clinical application.can save the medicinal material and the cost,creates the economical and the social benefit.
