Objective: To study the chemical constituents and quality evaluation of Sauropus rostratus Miq.And provide scientific basis for the utilization and development of S.rostratus.Methods: S.rostratus was extracted by ethanol,and separated into four parts according to the polarity,inc luding petroleum ether part,ethyl acetate part,n-butanol part and water part.Each portion was isolated by silica gel,macroporous resin adsorption,polyamide,Sephadex LH-20 chromatography,et al.The structures were identified by spectral analys is(MS,1HNMR,13 CNMR,HMBC,HMQC et al)and comparing with the published data.The active ingredien caffeic acid of S.rostratus was qualitatively identified by TLC,and quantitative dtermined by HPLC.Results: Twenty-five compounds were isolated and Twenty of them were identified as n-Triacontano(1),2,4-di-tert-butylphenol(2),?-sitosterol(3),daucosterol(4),glyceryl palmitate(5),3,6-Anhydro-2-deoxy-D-arabino-hexono-l,4-lactone(6),niacinamide(7),scopoletin(8),3?-hydroxy-26?,27?-arbor-13(18)-ene-24-carboxylic acid(9),2R,3R,5S-trihydroxy-6R-nonadecyltetrahydropyran-4-one(10),emodin(11),pr-otocatechuic acid(12),2,3-Dideoxy-D-erythro-hex-2-enono-1,4-lactone(13),caffeic acid(14),3-acetrlc-affeicac id(15),quercetin(16),D-galactose(17),mannitol(18),aurant iamide acetate(19),kaempferol(20).TLC identification for S.rostratus was highly specific.Caffeic acid content had a good linearity in the range of 0.024~0.6mg·m L-1(r=0.9999),the result of methodology validation meets the requirement.Conclusion: 3?-hydroxy-fernane-26?,28-dimethyl-13(18)-en-oic acid(9)is new compound,designated as Saurolic acid?Compound 2,5,6,7,8,9,10,11,12,13,14,16,17,18,19 are isolated from Sauropus Bl.for the first time.The established method can be used for the estimation of quality of S.rostratus.
