The Related Interaction And Significance Of Ubiquitin Degregation Between HRD1 And LAT1 In Gastric Cancer Cell

Objective: To investigate the expression of L-type amino acid transporter and the change of the proliferation,invasion and metastasis in gastric cancer cells with Regulating the expression of ubiquitin ligase HRD1,to explore the interaction and the significance.Method: si RNA and sh RNA were Disigne,synthesize and transfecte into SGC-7901 gastric cancer cells,examined the protein expression and m RNA of HRD1 and LAT with Western blot and q RT-PCR respectively.By using MTT assay,,Transwell chambers to explore the proliferation,migration.Results:The protein expression of LAT1 increased after downregulate the protein expression of HRD1,but the migration and proliferation of the SGC-7901 gastric cancer cell were promoted.Upregulation of HRD1 expression can inhibited the migration and proliferation,The protein expression of LAT1 decreased(P<0.05)Conclusions:The protein expression of LAT1 changed on the contrary after regulativing the HRD1.The ivasion,proliferation of the SGC-7901 gastric cancer cell change follow the protein xepression of LAT1.It suggests that HRD1 and LAT1 play an important role in the progression of SGC-7901 gastric cancer.
