Significance Of Ubiquitin Ligase Hrd1 And L-type Amino Acid Transporter-1 Expressed In Human Gastric Carcinoma

Objective:To investigate the expression of ubiquitin ligase hrd1 and L-type amino acid transporter 1 in gastric carcinoma and in normal cells,to explore their roles in the development of gastric carcinoma.Methods: Collect 63 cases of gastric carcinoma samples from Hospital,the control group was the tissues 5cm from the edge of the cancer.By using immunohistochemical SP method,Q-PCR and western-blot,research the expressions of hrd1 and LAT1 protein in 63 cases of normal human cells and gastric carcinoma cells.Results: 1.immunohistochemical results:Expression level of hrd1 protein was significantly higher in human gastric carcinoma cells than that in normal human cells(P<0.05). But not with histologic grade and lymph node metastasis.The expression of LAT1 was higher in human gastric carcinoma cells than that in normal human cells,stage Ⅲ-Ⅳ, tumor size≥5cm, were significantly higher than those in stage Ⅰ-Ⅱ, tumor size <5cm(P<0.05). 2.The results of Q-PCR:The gene expression was described by relative expression levels.The results showed that hrd1 and LAT1 gene has a higher level of expression in gastric carcinoma than that in normal tissues.The difference was statistically significant(P<0.05). 3.The results of western-blot:Measured the gray value of each group and their internal reference,the ratio of them is the relative gray value.The higher the value,the higher the gene expression,The results showed that the expression level of hrd1 and LAT1 protein was higher in gastric carcinoma than in normal tissues,the difference was statistically significant(P<0.05).4.The high expression of LAT1 was positive correlated to hrd1 in gastric carcinoma(P<0.001).Conclusion: The expression of hrd1 and LAT1 was higher in gastric carcinoma than that in normal human cells.It suggests that hrd1 and LAT1 may play an important role in the occurrence and progression of gastric carcinoma.
