Objective:To discuss the paraquat on the mouse brain damage and to detect the expression of inmunothistochemical antibodies about NGF and NOS in chronic paraquat poisoning rats' brain,we gave C57BL/6 rats some doses of paraquat by intraperitoneal injection.We could futher learn whether paraquat do some damage to brain and discuss the treatment of cyclosporine-A on brain damage.Methods:Thirty C57BL/6 rats with six-week size were divided into three groups randomly :10 rats for compare group,10 rats for experimental group one and the remaining 10 rats for experimental group two.The compare group was given 0.9% normal saline 10mg/Kg and the experimental group one and experimental group two were 20% paraquat 10mg/Kg by intraperitoneal injection,for twice a week,and last eight weeks at all.Then the experimental group two was given cyclosporine-A by intraperitoneal injection for therapy.During the process of building up the animal models,we observe rats' hair,movement,general conditions and behavioral changes in three groups successive,which includes water maze experiment,suspending experiment and automatic activities counting.All rats were putted to execution and the brain tissue were collected and cutted into slices for pathology research with inmumohistochemical study.Results:1.During the process of paraquat intraperitoneal injection,the C57BL/6 rats of group experimental one and experimental two appear the phenomenon with slightly tremble,action stiff,hair turn into yellow,motion discordant and the ability of avoding stimulate weaken.The compare group did not have obvious change.2.During the experiment of water maze and suspending,experimental group one and experimental group two have obvious change which contrast with compare group.3.The expression of inmunothistochemical antibodies NGF and NOS are obviously enhanced in experimental group one,which distributed in striatum and substantial nigra almostly.The experimental group two were less than experimental group one in the expression of NGF and NOS.4.The expression of compare group with inmunothistochemical antibodies NGF and NOS were less than experimental group one and experimental group two.Conclusion:1.Exposed the cricumstance of paraquat,the rat would appear the behaviour of Parkinson's disease,such as slightly tremble,action stiff,hair turn into yellow,motion discordant and the ability of avoding stimulate weaken.Then we could infer paraquat had something to do with rat brain damage and paraquat could induce PD animal models.2.The expression of experimental group one and experimental group two with NGF and NOS enhanced in substantial nigra mainly which could deduced have some relationship with PD.3.The experimental group two by cyclosporine-A intraperitoneal injection which express NGF and NOS less than experimental one had some function on rat brain damage.However,the behaviour of two groups did not has distinct change.
