Objective: Eczema is caused by a variety of internal and external factors having a significant leakage tendency of inflammatory skin disease.The disease is common dermatology,China's general population prevalence rate is about 7.5%,US 10.7%[1?2].Which is characterized by the acute phase lesions with erythema,high herpes mainly exudation tendency;chronic phase lichenification based,easy to recurrent disease,severe itching,seriously affecting the quality of life of patients,has become the field of dermatology hotspot.But eczema pathogenesis is not clear,studies confirm,TH1 / TH2 differentiation imbalance is one of the main mechanisms of pathogenesis of eczema,TSLP and IL-10 will play a pivotal role in the Thl and Th2 balance.TSLP is an important factor to start allergic inflammatory diseases,and in a variety of allergic diseases(atopic dermatitis,asthma,etc.)can be detected increased levels of TSLP expression[42~43].TSLP activated DCs induced by the original Th0 cells into TH2 transformation,leading to Th2 cytokines(IL-4,IL-5,IL-13)and raised levels of TNF expression by TSLP-DCs activated CD4 + T cells can reduce IL the expression levels of 10 and IFN-?[3],the Two cytokines may reduce inflammation caused by Th2 cells[4].IL-10 inhibits monocyte-macrophages promote natural and specific immune function,limitations and prevent excessive immune responses;induce peripheral tolerance when the antigen persists in the body to enhance the immune system of allergy scavenger function[5].Currently,TSLP and IL-10 Factors in eczema patients less chronic eczema transition mechanism from acute phase inflammatory TH2 led to the TH1 response to inflammation mainly unclear.Therefore,further research TSLP and IL-10 expression and its clinical significance factor in eczema patients,might clarify the pathogenesis of eczema and explore new therapeutic targets play a positive role,and for the TSLP and IL-10 study in allergic diseases provide some reference.This topic from the following several aspects to discuss56:1 TSLP,IL-10 expression differences between patient and normal eczema.2 Eczema patients at different stages of TSLP,IL-10 expression differences.3 TSLP eczema in patients with serum IL-10 expression levels of correlation.4 TSLP,IL-10 expression and EASI score,itching intensity correlation.5 Eczema patients with different gender TSLP,IL-10 expression level differences.6 TSLP,IL-10 expression correlation between the level of sick time.Method:1 Referring to debate editor Zhao clinical dermatology diagnostic standards and related documents selected 60 cases of eczema patients,34 males and 26 females,aged 1-74 years(mean 36.38 20.88 disabilities)years,"according to the clinical manifestations of the patient staging,of which 20 cases of patients with acute,subacute phase 20 cases,20 cases of chronic phase;20 cases of healthy persons(7 males and 13 females,aged 13-44 years,mean 29.45 years old with disabilities 8.34)as a control group.2 Sterile 3mL peripheral venous blood drawn join ordinary drying tube and allowed to stand 30 min centrifugation(3000r/min,l0min)natural state,isolated serum,-80?low-temperature freezer frozen under test,and collected clinical data of patients with eczema,according to EASI scoring criteria for 60 cases of eczema score.3 Solid phase sandwich enzyme-linked immunosorbent assay(ELISA)method,in strict accordance with the kit instructions requirements,10 IL-concentration detecting TSLP and specimens.Analysis of TSLP,IL-10 between patients and normal eczema,different stages,expression of gender differences and eczema patients with EASI score,itching and other clinical indicators of the strength of the correlation between the use of statistical methods.Results:1 Acute and sub-acute phase serum concentration group eczema TSLP is 39.03 persons 12.92pg/mL,the expression level was significantly higher than in chronic phase(concentration of 22.53 persons 9.43pg/mL)and normal control group(concentration disabilities 5.81 19.71pg/mL)(P<0.05),and TSLP levels in acute,subacute phase(concentration of 30.78 persons 9.24 pg / mL)and chronic group(P<0.05)difference between the groups was statistically significant,but TSLP concentration level was not statistically different between the chronic phase and normal group.2 Subacute eczema patients serum concentrations of IL-10 10.26 persons 3.61Pg/mL,the expression level was significantly higher than the acute phase(concentration of 7.16 persons 2.32pg/mL),patients with chronic phase(concentration of 6.42 persons 1.42pg/mL)and normal control group(concentration of 5.92 persons 2.42pg/mL)(P<0.05),but the expression of IL-10 level was not statistically different between the acute phase,chronic phase and normal group.3 TSLP expression in patients with eczema serum IL-10 levels were not significantly correlated,but the serum of patients with subacute phase eczema 10 IL-TSLP expression level was negatively linear correlation(r=-0.582,P <0.05).4 Eczema patients serum levels of TSLP expression EASI score positive linear correlation(r = 0.491,P<0.05),IL-10 expression and EASI score was no significant correlation.5 TSLP expression in the serum of patients with eczema between linear and pruritus intensity levels positively correlated(r = 0.308,P<0.05),whereas no significant correlation between IL-10 levels and pruritus intensity.6 TSLP,IL-10 expression levels and eczema disease duration,gender,erythema was no significant correlation between grade;however,the degree of erythema grouping of moderate and severe groups TSLP expression was significantly higher than mild erythema and no group(P<0.05),erythema the extent of each packet between IL-10 expression was no significant difference in levels.7 Eczema patients serum TSLP expression was positive linear correlation between the level and the size(r=0.490,P<0.05),whereas no significant correlation between IL-10 levels and the size.8 There was no significant difference TSLP,IL-10 expression levels between patients with different gender eczema.Conclusion:1 TSLP expression in the serum levels were significantly higher in patients with eczema,with a statistically significant difference,IL-10 expression levels increased in the serum of patients with eczema,and the sub-acute phase is more obvious,with statistical difference,suggesting that these two cytokines may play an important role in their pathogenesis,and have some correlation with the severity of the patient's disease.2 Eczema subacute phase serum levels of IL-10 expression was significantly higher than the acute,chronic patients and the control group(P <0.05),eczema and sub-acute phase serum TSLP and IL-10 expression was a negative linear correlation between level(r=-0.582,P<0.05),prompted IL-10 expression of TSLP may have a certain extent,may act as a disincentive in the pathogenesis of eczema,Transforming the process should be presumed chronic inflammatory response in TH1 mainly to play a role in anti-inflammatory TH2-based acute phase.3 Eczema patients serum TSLP expression and EASI,pruritus intensity,size was positively correlated,can determine the activity and severity of the patient's condition index eczema,TSLP could be a new factor causing itching may occur in pruritus mechanism plays an important role.
