Objective To study the effect of a new traditional Chinese medicine named Bushen Liaogeng Extract on serum sex hormone, hypothalamus monoamine neurotrasmitters, hypothalamic ?-endorphin, free radical metabolism, organizational structure of uterus of rats model of menopause, and to study the effect of Bushen Liaogeng Extract on serum sex hormone, follicular reserve, granulosa cell apoptosis,organizational structure of uterus and ovaries of the aged mice and to explore the mechanism.Methods(1) Menopause models were established by surgical removal of the bilateral ovaries of female SD rats. The rats were ig administered once daily for 4weeks. Abdominal aorta blood was taken to measure serum estradiol(E2), luteinzing hormone(LH), follicle stimulating hormone(FSH), gonadotropin releasing hormone(Gn RH), testosterone(T), malondialdehyde(MDA) content and superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) activity using enzyme-linked immunosorbant assay(ELISA) method,and hypothalamus was taken to measure dopamine(DA), norepinephrine(NE), 5-hydroxytryptamine(5-HT), 5-hydroxyindole acetic acid(5-HIAA), ?-endorphin(?-EP) content using BCA protein qualification and enzyme-linked immunosorbant assay(ELISA) method. Bilateral uterus were sampled for the calculation of organ index and endometrial thickness and for the observation of pathological changes.(2) Menopause models were established by using aged mice.The mice were ig administered once daily for 4 weeks.Eye canthus blood was taken to measure serum estradiol(E2), luteinzing hormone(LH), follicle stimulating hormone(FSH),progesterone(P) content using enzyme-linked immunosorbant assay(ELISA)method.Bilateral ovaries were sampled for the calculation of organ index, the number of ovarian follicle and corpus luteum, the observation of pathological changes,and the number of apoptotic mouse ovarian granulocytes was detected by DNA in situ end labeling(TUNEL) method. Bilateral uterus were sampled for the calculation of organ index and endometrial thickness and for the observation of pathological changes.Results(1)Bushen Liaogeng Extract can effectively increase serum E2 content(P<0.05,P<0.01), lower LH, FSH, Gn RH, T, MDA contents(P<0.05,P<0.01), increase SOD, GSH-Px activity(P<0.05,P<0.01), lower hypothalamus 5-HT, 5-HIAA, DA, NE contents(P<0.05,P<0.01), increase hypothalamus ?-EP content(P<0.05), thicken the endometrial thickness(P<0.05), elevate the uterus index(P<0.05), and improve the ultrastructure of the uterus and ovaries in aged mice.(2) Bushen Liaogeng Extract can effectively increase serum E2 and P content(P<0.05,P<0.01), lower FSH and LH contents(P<0.05,P<0.01), increase the number of ovarian follicles at various levels(P<0.05,P<0.01), reduce ovarian granulosa cell apoptosis(P<0.01), thicken the endomerial thickness(P<0.05,P<0.01), elevate the uterus and ovary indexes(P<0.05,P<0.01), and improve the ultrastructure of the uterus and ovaries in aged mice.Conclusion Bushen Liaogeng Extract have effectively function on menopause syndrome, it's mechanisms are as follows: ? Cure menstrual disorders, irregular menstruation, frigidity and other symptoms of menopause syndrome by regulating serum sex hormone, hypothalamus-pituitary-ovary axis function and endocrine. ? Cure insomnia, restlessness, irritability and other symptoms of menopause syndrome by regulating contents of monoamine neurotransmitters in hypothalamus. ? Cure menstrual cycle disorders, emotional disorders, hot flashes and other symptoms of menopause syndrome by regulating content of endogenous opioid peptide in hypothalamus and the function of hypothalamic endocrine and body temperature regulation.(4) Delay the menopause process of tissue oxidative aging and organ failure by regulating peroxidase activity in blood and the contents of free radical metabolites.(5) Enhance ovarian reserve by regulating the number of ovarian follicle and luteum.(6) Delay the process of menopause ovarian failure by inhibiting ovarian granulosa cell apoptosis.(7)Improve the reproductive function by improving the ultrastructure of the uterus and ovaries.
