| Objective: To determine the effect of polycystic ovary-like environment and insulin sensitizer, rosiglitazone on secretion of IGF-2, PAPP-A, E2, P from human granulosa luteal cells in vitro. Methods: Granulosa cells from infertility women undergoing IVF-ET with normal ovulation were cultured in serum-free medium. According to the additive, the granlosa cells of each patient was divided into five groups: androstenedione(A) group, LH group,insulin(INS) group, A+LH+INS group, and A+LH+INS + rosiglitazone(R) group, and each group was subdivided into two subgroups with high (polycystic ovary-like environment) or normal (physiological environment) level of additive. After cultured for three days, the concentration of IGF-2, PAPP-A, E2, P in cell-conditioned medium of each group were detected. Results: The concentration of IGF-2, PAPP-A in polycystic ovary-like environment were decreased in A group and INS group (P>0.05), and increased in LH group and A+LH+INS group (P<0.01); Concentration of IGF-2, PAPP-A in LH group and A+LH+INS group were all higher than that in A group and INS group (P<0.01), with a little reduce in A+LH+INS group comparing to LH group and no difference between A group and INS group (P>0.05 respectively). As concern as secretion of E2, P, the concentration of E2, P in polycystic ovary-like environment were significantly decreased in A group and A+LH+INS group, and significantly increased in LH group and INS group (P<0.01 respectively); Concentration of E2, P of LH group, INS group and A+LH+INS group were all significantly higher than that of A group, with the highest level of E2 in INS group and the highest level of P in LH group; Moreover, the concentration of E2 in A+LH+INS group compared to INS group and the concentration of P in A+LH+INS group compared to LH group were both significantly decreased in polycystic ovary-like environment , but no difference were found in physiological environment. Concentrations of E2, P of A+LH+INS group were significantly decreased after adding rosiglitazone, but no differences were found as to IGF-2, PAPP-A. Conclusion(s): In polycystic ovary-like environment in vitro, A could possibly inhibit not only basic but INS or LH stimulated secretion of E2, P from normal granulosa luteal cells; In polycystic ovary-like environment in vitro, secretion of IGF-2, PAPP-A from normal granulosa luteal cells could possible be promoted by LH, but this promoted effect could possible be inhibited by A, INS. Rosiglitazone maybe could inhibit secretion of E2, P from normal granulosa luteal cells, but had not effect on secretion of IGF-2, PAPP-A. |
