In living body,autophagy which makes long-lived proteins and organelles access to catabolic processes for cell reuse is ubiquitous.Lower levels of a certain degree of autophagic activity which participate in the adaptive immune response is a self-protection mechanism.Insufficient or excessive autophagy will leads cells appear homeostasis and increased cell damage,even cause cell death.Existing researches show that the environment and pathological stimuli will increased autophagic activity in different degrees,the process is more closely related to the development and progression of heart disease.Previous research of our laboratory discovered that different exercise on the heart exert different regulation.Low levels of exercise training can improve the function of cardiac.However when the movement is performed at high intensity overload,rats will attain sports fatigue and will produce severe damage to cardiac function.Thus,the regulation of cell autophagy activity is an important potential target for the treatment of cardiovascular diseases.Thus,we believe that autophagy activity may play an important role in the movement of fatigue induced myocardial injury in the process.The purpose of this experiment is to explore the rat cardiomyocyte autophagy level in sports fatigue and training change and its molecular mechanism.Swimming endurance training methods was used to built sports fatigue rats model.Frozen sections,electron microscopy was used to object the cardiac morphology and histology of rat.DCFH-DA probe was used to detect the ROS that observed in different exercise intensity rats.By Western Blot detect the level of autophagy.Results of the study are as follows.The myocardial of fatigue rat has became compensatory hypertrophy and was seriously damaged.ROS and autophagy levels of fatigue rat were significantly improved.When NAC was injected,autophagy level of fatigue rat occurred significantly reduced.The myocardial of exercise rat has became increased.But myocardial cell structure is still intact.Serious damage has not occurred.There results illustrate: Sports interference raise the level of autophagy and ROS in rat cardiomyocytes.When fatigue occur,excessive autophagy destroys the cell structure,which is related by ROS signaling pathway.The research provides an important theoretical basis for the prevention of fatigue and cure the cardiac diseases caused by motion.
