1 ObjectiveObservation of Electroacupuncture on spinal cord ischemia again after reperfusing injury mediated endothelin 1(IL-1), c-Jun N-terminal kinase(JNK)activator protein-1(AP-1) index changes, through IL-1/JNK/AP-1 signaling pathway of Electroacupuncture on spinal cord ischemia reperfusion injury in rabbit spinal cord protection of molecular regulation mechanism.2 Methods24 Chinese white rabbits were divided into sham operation group, model group and EA group, with 8 rats in each group. Using zivin improvement method to duplicate the animal model of rabbit SCII. electroacupuncture group induce Yanglingquan,hanging bell(bilateral)in 30 min treatment before building a model of electro acupuncture, model group do acupuncture and sham operation group do not clip occlusion of the abdominal aorta. Before ischemia, take rabbit femoral vein blood 2ml after 1h, 4h, 8h, 12 h reperfusion and animals were sacrificed after 12 h reperfusion, and then take out lumbar(L2-L5) spinal cord tissue, ELISA method was used to detect the content of IL-1, IL-1Ra, JNK, AP-1, HE staining method to observe the pathological morphology of the spinal cord, immunohistochemical method, blot Western detection of lumbar spinal cord(L2-L5) spinal cord JNK, AP-1 protein expression, and semi quantitative analysis.3 ResultsSerum levels of IL-1 beta in Model group compared a very significant difference(P < 0.01) with sham operation group at the same time. Electroacupuncture group, the serum levels of IL-1 beta and model group(group B) three time points(reperfusion 4h,8h, 12h) have significant difference(P < 0.05), which after the reperfusion 8h, 12 h hasvery significant difference(P< 0.01). Serum IL-1ra in model group and sham operation group the same time compared with very significant difference(P< 0.01); electro acupuncture serum IL-1ra and model group at the 3 time points(reperfusion for 1h, 4h,12h) compared with significant difference(P < 0.05).In model group, the serum levels of JNK and sham operation group at the same time there was significantly increased(P < 0.01); Electroacupuncture group, the serum levels of JNK and model 4 time points group(reperfusion for 1h, 4h, 8h, 12h) had significantly decreased(P < 0.05); in the model group, the spinal cord ischemia and12 h after reperfusion in rabbit spinal cord nerve cell cytoplasm and nucleus of JNK is abundantly expressed, electroacupuncture group in a JNK by cytoplasmic to nuclear translocation, a lot of detention in the cytoplasm; spinal cord of rabbits in the model group JNK3 mean relative expression of quantity increased significantly(P < 0.01),electroacupuncture group rabbit spinal cord JNK3 mean relative expression of lower amount of model group decreased significantly(P < 0.01).In model group, the serum levels of AP-1 increased significantly compared with sham operation group at the same time(P < 0.01);Comparison of the serum levels of AP-1 between electroacupuncture group and model group, significant differences(P<0.05) appear after 1h, 8h reperfusion; in the model group, the spinal cord ischemia and 12 h after reperfusion rabbit spinal cord nerve cells in the nucleus of AP-1have large expression, electroacupuncture group in the spinal cord ischemia again 12 h after reperfusion AP-1 in the nucleus decreased expression of; AP-1 spinal cord of rabbits in the model group the average relative expression quantity compared with the sham operation group increased significantly(P < 0.01); Electroacupuncture group rabbit spinal cord AP-1 mean relative expression of lower amount of model group decreased significantly(P < 0.01).4 Conclusion1 rabbit SCII can cause an increase in rabbit serum IL-1, IL-1Ra, JNK, AP-1, JNK and AP-1 protein expression were significantly increased in the lumbar spinal cordtissue.2 electro acupuncture can down regulate the content of IL-1, JNK, AP-1, SCII,increase the content of IL-1Ra, and reduce the expression of JNK, AP-1 protein in the spinal cord tissue of the lumbar spinal cord of the rabbit.3 electroacupuncture through the IL-1/JNK/AP-1 signaling pathway and play a protective effect on the spinal nerve function of SCII rabbits.
