1.ObjectiveBy observing changes in serum and spinal cord tissue in IL-1,JNK,AP-1 index,spinal cord tissue morphology,demonstrated that the spinal cord ischemia reperfusion and abnormal activation of IL-1/JNK/AP-1 pathway is closely related to clear IL-1/JNK/AP-1 pathway may be an important approach to electroacupuncture treatment.Which is an effective method for the treatment of spinal cord ischemia reperfusion injury,which is of great significance.2.Methods32 New Zealand rabbits were randomly divided into model group,EA group,SP600125 group and SP600125+ EA group.Each group has 8 rabits.The modified Zivin method was used to establish the model of spinal cord ischemia-reperfusion injury in rabbits,and the rabbit model was established before one hour.EA group:Electroacupuncture of "Yanglingquan" and "Xuanzhong" point;SP600125 group: rabbit spinal cord ischemia reperfusion injury model was established by 30 minutes ago.SP600125+ electro acupuncture group: SP600125 treatment + electroacupuncture treatment group.Model group: Do not inject any drugs and electro acupuncture.After the establishment of the model of spinal cord ischemia and reperfusion in rabbits and 30 minutes after spinal cord ischemia,The L2-L5 spinal cord was taken out when examination was finished.The levels of IL-1?,IL-1Ra,JNK and AP-1 in serum were detected by ELISA.The expression of IL-1?,JNK and AP-1 were detected by immunohistochemistry.Quantitative analysis of JNK and AP-1 in injured spinal cord were detected by Western blot.3.Results1.Model group: reducing the number of neurons,neuronal damage,unclear structure,visible nuclear condensation or disappear,close to the edge of the cell nucleolus,nuclear membrane damage,Nissl bodies appear not clear,focal hemorrhage,some apoptotic bodies,a small amount of neutrophil infiltration,vacuolization around beta,IL-1Ra,IL-1;JNK and AP-1 at different time points in rabbit serum were increased(P<0.05).IL-1,JNK and AP-1 expression in spinal cord tissue.2.Comparison of EA group,SP600125 group,SP600125+ EA group and model group of spinal cord tissue morphology: EA group,SP600125+ EA group,SP600125 spinal cord tissue pathology was improved,the number of apoptotic neurons decreased;but both of neurons in the spinal cord tissue showed different degrees of bleeding,inflammatory cell infiltration;electro acupuncture group,SP600125 group,SP600125+EA group compared with the model group,the damage of spinal cord neurons is light,the number of normal neurons increased obviously than the model group(P<0.05).3.Comparison of EA group,SP600125 group,SP600125+ EA group and model group IL-1 beta and JNK and AP-1 contents: Electroacupuncture group,SP600125 group,SP600125+ EA group,SP600125+ EA group in addition to the 4H time point in IL-1 beta increased slightly,while the serum levels of IL-1,JNK and AP-1 expression were significantly reduced,in the model group was lower than that of IL-1,JNK,beta AP-1,was statistically significant compared with the model group(P<0.05),and electroacupuncture and SP600125 expression in the same trend;EA group,SP600125 group and model group of IL-1Ra reperfusion 4H were statistically significant(P<0.05),the mean IL-1Ra was increased only in the SP600125+ group;8h and model group at the same time the difference was statistically significant(P<0.05),but the mean IL-1Ra is reduced.4.Conclusion1.The experimental data show that: when the spinal cord ischemia reperfusion injury occurs in rabbits,IL-1?,IL-1Ra,JNK and AP-1 levels in serum will increase.2.The intervention effect of electroacupuncture and SP60012 on ischemia reperfusion injury of the spinal cord,by promoting the increase of IL-1Ra,Lastly,IL-1 ? JNK and AP-1 was restrained.Therefore they protect the spinal cord caused by ischemia reperfusion injury of nerve function damage.3.The change trend of the EA group and SP600125 group IL-1 ? and JNK and AP-1 were tsame,suggesting that neuronal damage and SP600125 electroacupuncture on spinal cord ischemia reperfusion induced similar protective effect,its mechanism may be through down-regulation of JNK and AP-1 activity,and decrease the content of IL-1?,reduce the number of neural cell apoptosis after ischemia reperfusion,so as to protect the spinal cord neurons.4.Electro acupuncture can reduce the expression of IL-1?,JNK and AP-1 by inhibiting the activation of IL-1/JNK/AP-1 pathway,and then inhibit the apoptosis of spinal cord neurons after spinal cord ischemia reperfusion.
