Objective:To observe the effect of Yanghe Pingchuan on the asthma model rats with chronic inflammation,the proportion of CD4+CD25+Tregs and SDF-1,and to probe the immune mechanism of bronchial asthma attack.Methods:Extract all the marrow of juvenile male SD rats by adherent wall to cultivate BMSCs,to amplify and to passage, vein input SD rats. Choose clean level 120 juvenile male SD rats, weight about 100±20 grams,the average random divided into eight groups,each group has 10 rats,including the normal group,the asthma model group, the BMSCs group,the BMSC swipe off Tregs group,the BMSCs wipe off Tregs Yanghe Pingchuan treatment group,the BMSC slow dose Yanghe Pingchuan treatment group,the BMSC high dose Yanghe Pingchuan treatment group and the BMSC Dexamethasone treatment group. Asthma model preparation using OVA and the Aluminum Hydroxide Gel to have sensitization and atomization, through tail vein injection of BMSCs suspension liquid and Monocloal Antibody CD25,extract the rats tail vein blood,adopting Yanghe Pingchuan,Normal Saline and Dexamethasone intervent the corresponding groups of rats. After HE staining microscopically observed groups of SD rats lung tissue pathology,Enzyme-linked Immunosorbent Assay(ELISA) to detect Bronchial Alveolar Lavage Fluid(BALF) in inflammatory cells count,protein immunoblot method(Western blot)and PCR detect groups of SD rats lung tissue SDF- 1 m RNA and protein expression.Results:1 Under electron microscope observe, BMSCs which were cultivated by whole bone marrow adherent wall method are long spindle, high purity, arranged spirally or radially, the proliferation ability is strong,and multi-directional differentiation potential, identified by FCM surface sign CD29, CD44, CD11 b, CD45 were 98.68%, 99.13%, 0.28%, 99.13%,and through the corresponding drugs can induce for osteogenesis and into fat cells, conform to the requirements of the international association of cell therapy on BMSCs identification.2 Rats compared with normal group,model group rats lung biopsy in inflammatory cells infiltration in great quantities, secretion increased, sputum bolt formed and blocked the airway, airway got narrow the total amount of inflammatory cells in BALF and various types of inflammatory cells, serum IL- 4 and IL-5 were significantly increased(P<0.01);Compared with the model group rats, Yanghe Pingchuan intervention group and dexamethasone group rats lung tissue pathological inflammatory cells, secretion and airway got stenosis,inflammatory cells in BALF, serum IL- 4 and IL- 5 drops were significant(P<0.05);And Yanghe Pingchuan for both high and low dose group dexamethasone group every two comparative were no difference(P>0.05).3 Compared with the normal group,the model group rats CD4+ CD25+Treg ratio decreased significantly(P<0.01);Compared with the BMSCs group, the BMSCs got rid of Tregs group declined significantly(P<0.01);Compared with the BMSCs got rid of Tregs group,the Yanghe Pingchuan intervention groups and the Dexamethasone group were declined obviously(P<0.01).4 Compared with the normal group,the model group rats SDF- 1 protein and m RNA expression quantity increased obviously(P<0.01); Compared with the model group,the BMSCsgroup,the Yanghe Pingchuan treatment groups and the Dexamethasone group rats SDF-1 protein and m RNA expression were decreased significantly(P<0.01).
