| Objective To establish mice asthma model of airway remodeling and investigate the potential suppression role of Pingchuan-Guben Decoction in airway chronic inflammation and remodeling by observing the expression of NF-κB and vascular endothelial growth factor(VEGF)on the mice model of asthma,further to provide new expedmental evidence forthe prevention and treatment ofairway remodeling in asthma.Methods Fifty Kunming mice were randomly divided into five groups averagely:negative control group(A),asthma model group(B),Pingchuan-Guben Decoction group(C),Budesonide group(D),Pingchuan-Guben Decoction+Budesonide group(E).Bã€Cã€Dã€E groups mice chronic asthmatic model of airway remodeling Was sensitized by OVA and aluminum hydroxide[Al2(OH)3],challenged by an aerosol of10%OVA.Total and differential cells were counted from bronchoalveolar lavage fluid(BALF),histologic examination were performed to observe the general pathologic alteration and analyze the thickness of airway wall.The protein expression of NF-κB and VEGF were calculated by immunohistochemistry.Data were evaluated using One-way ANOVA,followed by least-significant difference(LSD)test.Results:â‘ There Was no wheezing symptoms in group A while wheezing in other groups.And group B were more serious than the treatment groups.There was no significant different between the treatment groups;â‘¡HE staining showed that compared with the treatment groups,there were a large number of inflammatory cells infiltration,heavier smooth muscle hypertrophy and mucous membrane hyperemia in the asthma group.â‘¢The inflammatory cells in BALF in asthma group B were significantly inereased than group Aã€Cã€Dã€E(P<0.01),And group A was significantly decreased than group Bã€Cã€Dã€E(P<0.01)The number of EOS was no remarkable difference between group C and D(P>0.01).â‘£The thickness of airway in each group was(6.61±1.14,16.66±1.52,11.57±1.26,9.53±1.93,8.56±1.35)μ2/μm respectively,F=54.59,P<0.01.airway thickness in group Bã€Cã€Dã€E were all significantly higher than group A(P=0.000,0.000,0.000,0.012respectively),airway thickness in group C also showed remarkably thicker than group D and E(P=0.009,0.000respectively),there was no differct between group D and E(P=0.193).⑤The VEGF mean IOD values(11.57±1.64,35.87±4.92,28.28±2.02,25.06±2.58,16.31±2.41respectively F=85.45,P<0.01)of mice lung in asthmatic model,group B was significantly higher than those in normal control group A and difference intervention group Cã€Dã€E(P<0.01).group A was significantly lower than those asthmatic model group B and difference intervention group Cã€Dã€E(P<0.01),group C was higher than group D and E(P=0.036,0.000respectively).â‘¥The NF-κB mean IOD values(16.52±2.04,20.88±0.78,19.57±1.21,18.32±0.93,17.66±0.78,respectively F=14.73,P<0.01)of mice lung in asthmatic model,group B was significantly higher than those in normal control group A and difference intervention group Cã€Dã€E(P=0.000,0.042,0.000).group A was significantly lower than those asthmatic model group B and difference intervention group Cã€D(P<0.01),There Was no significant difference between group A and E,group C and D(P=0.76,0.53.).Conclusions:Sensitization and aerosol of OVA can successfully establish chronic mice asthmatic model of airway remodeling;The expression of VEGFã€NF-κB all increase conspicuously in asthma,which could promote the progress of airway inflammation and remodeling.Pingchuan-Guben Decoction could be down NF-κB and VEGF expression of asthma on airway inhibit airway chronic inflammation and remodeling. |
