Objective:This study aimed to observe the neurological deficits? morphology surrounding hematoma tissue,AQP-4 expression,the different period of brain water content after using atorvastatin among acute(ICH)in rats,understanding AQP-4 expression under the effects of atorvastatin,speculated the relationship among AQP-4 content,cerebral edema,discuss whether atorvastatin could protect nerve cell after intracerebral hemorrhage,prospect to offernew guide for ICH treatment.Methods: According to the random number table method,65 male SD rats were fellinto 3 groups: atorvastatin treamtment group(AT gropup,n=25),intracerebral hemorrhage group(ICH group,n=25),no treatment control group(NC,group,n=15),each group was averagely fell into five subgroups as 6h,1d,3d,5d,7d.In ICH group,using the method of modified two times injection of 50 ul autologous arterial blood which is not solidified.Equivalent saline take the place of autologous arterial blood in NC group.AT groups were given atrovastin(10mg/kg/d)for seven days.As comparing research,other rats were given the same amount of saline.After the success of modeling,the rats at each time point were collected for neurological dysfunction score named Garcia.Water contents of a part brain tissue arounding the hematoma were observed at different time points by wet-dry-weight method,the other of rat brain was saved in 4% neutral Formaldehyde Solution,for paraffin embedding,slicing,staining for HE,AQP-4 for immunohistochemistry.Result:(1)?The neurological deficits score of AT group and ICH group is lower compared with NC group at each time point;the score of AT group contrast to ICH group is higher at 1d ?3d?5d?7d group;Garcia's score was lowest at 3d groupwhether in ICH group or AT group,had statistics obviously(P<0.05).(2)?Changes of brain tissue morphology surrounding hematoma: structure of brain tissue was complete in NC group.Nerve cells around the hematoma were edematous of AT group and ICH group,a large number of red blood cells and a few inflammatory cells were visible at 6h,1d.we can see necrosis,nuclear condensation,red blood cells decrease,neutrophil infiltration at 3d group.The edema of brain neurons and inflammatory cells were obviously decreased whereas abound proliferation of glial cells at 7d group.Changes of brain tissue morphology of AT group was resemble to ICH group at 6h,1d,whereas the swelling nerve cells were less than the ICH group and we can observe a few inflammation cell infiltration and proliferation of glial cells at 3d,7d yet.(3)The water content of brain tissue arounding brain hematoma of ICH group and AT group were much more than the NC group,the 3d subgroup in ICH group reached the peak,had statistic obviously(p<0.05)?The water content showed lower level in AT group than ICH group at 1d?3d?5d?7d subgroup.(4)?In ICH group and AT group,the expression of AQP-4 cells analyzed by immunohistochemistry were increased at 6h,obviously increased at 1d,and reached the peak at 3d,had statistics obviously(P<0.05),started to decrease at 5d and could be observed at 7d yet.There is much more AQP-4 cells in AT group and ICH group than NC group,which could be found express much more in ICH group than AT group at any time.(5)? Linear correlation: The neurological dysfunction of ICH subgroups was nagatively correlated with the water content of brain tissue arounding brain hematoma and AQP-4(R2=-0.681,R2=-0.780,P<0.05).The AQP-4 level of ICH group were positively relatedto water content of brain tissue(R2=0.950,P <0.05).Conclusions:(1)We can found obviously pathological changes on brain structure after ICH and brain wherewater content and AQP-4 expression rised meanwhile,the expression of AQP-4 and brain edema is related to the severity(2)atorvastatin can improve nerve function,reduce brain edema,drop the expression of AQP-4 after ICH.
