| ObjectiveAn ICH (intracerebral hemorrhage) model of rats was established through adopting stereotaxic surgery and injecting the collagenase VII into the caudate nucleus of rats. Changes of brain water content and NgR(nogo recept) expression around the hematoma were assessed and compared between rats of ICH and ICH rats intervened by edaravone and atorvastatin respectively to study the neuroprotective effects of edaravone and atorvastatin in the therapy of ICH.MethodsFirstly, 160 Sprague-Dawley rats were divided into four groups at random: sham-operative group (Group A), ICH model group (Group B), edaraven therapy group (Group C) and atorvastatin therapy group (Group D), each of which had 40 rats. In Group B, Group C and Group D, ICH models were established through stereotaxically injecting 2μl saline and 0.5u collagenase VII into right caudate nucleus by a micro-infusion pump, while the sham-operative group only received 2μl saline. From 6 hours after operation on, 6mg/kg edaravone was injected by intraperitoneal injection into rats of group C once per day from the 1st day to the sacrifice day, and 10mg/Kg atorvastation was injected by intragastric injection into rats of group D once per day from the 1st day to the sacrifice day. Secondly, each group was divided into two sub-groups (Sub-group a & Sub-group b). Sub-group a contained 15 rats and was divided into three branch groups on average, which were killed on 3rd, 7th, 14th day respectively, and then right side cerebra of each branch group of Sub-group a were measured by dry-wet weight method to get the brain water content. Sub-group b contained 25 rats and was divided into five branch groups on average, which were killed after evaluating neurological symptoms by Forelimb Placing Test and Longa Test on 3rd, 7th, 14th, 21st and 28th day respectively, and then the brain tissues around the hematoma of each branch group of Sub-group b was examined by immunehistochemical method to observe the expression of NogoR. ResultsCompared with the sham-operative group, the neurological defects were more severe on each point in ICH group group(P<0.05), the brain water content was obviously higher in ICH group on 3rd and 7th day(P<0.01), and no difference on 14th day(P>0.05). The neurological deficit score has significantly improved in edaraven therapy group and atorvastatin therapy group than that in ICH model group on 3rd,7th,14th,21st and 28th day (p<0.05, p<0.05, p<0.05, p<0.05, p<0.05). The water content in brain tissue was significantly lower in ederaven therapy group and atorvastatin therapy group than that in ICH group on 3rd and 7th day(p<0.01, p<0.01), there was no significant difference on 14th day(p>0.05).In sham-opertive group the NgR expression was slight. In ICH model group, edaraven therapy group and atorvstatin therapy group, the NgR expression was obvious, and increased on 3rd day, reached a peak on 21st day, then discreased, but was still higher than that in sham-operative group on 28th day(P<0.05). Compared with the ICH group, the expression of NgR in the edaraven therapy group and atorvastatin therapy group obviously decreased(P<0.05).ConclusionsEderavon and atorvastatin can promote the regeneration of neurone in rats after ICH. It maybe achieved by reducing brain water content and discreasing the expression of NgR. |
