Objective:Genetic factors play an important role in the development of type 2 diabetes mellitus(T2DM), obesity and metabolic syndrome (MS). Objective of this study was to examine the associations of rs320 and rs285 polymorphisms in LPL gene with T2DM, obesity and MS by a case-control study and Meta-analyses.Methods:A totalof 1225 subjects of Hubei Han Chinese from Yichang, Hubei province were recruited. Three groups of case-controls were designed and genotyped by restriction fragment length polymorphism (RFLP). Chi-square test, Logistic regression and Meta-analyses were used to investigate the relationship between LPL rs320 (intron 8)/rs285 (intron 6) polymorphisms and T2DM, obesity and MS. Association strength is calculated by total odds ratio (OR) and 95% confidence interval (CI).Results:1. T2DM:Logistic regression analysis showed no association with T2DM for rs320 and rs285. Meta-analyses indicated an association with T2DM for the rs285 CT genotype (OR=1.30, P=0.049) in this study included 21 study data.2. Obesity:Logistic regression analysis with and without adjustment showed no association for rs320 and rs285 in three models. Meta-analyses revealed an association with obesity for the rs320 GG+TG genotype (OR=2.34, P=0.015) in this study included 6 study data.3. MS:Logistic regression analysis with and without adjustment showed no association for rs320 and rs285. Meta-analyses showed an association with MS for the rs320 GG+TG genotype (OR=0.82, P=0.048) and rs285 CT genotype (OR=1.32, P=0.017) in this study included 9 study data.Conclusions:1. LPL rs320 and rs285 polymorphisms were not associated with T2DM in Hubei Han Chinese. Meta-analyses indicated an association between LPL rs285 CT genotype and T2DM.2. There is no association between LPL rs320/rs285 and obesity in Hubei Han Chinese. Meta-analyses indicated that rs320 GG+TG genotype increases the risk of obesity.3. There is no association between LPL rs320/rs285 and MS in Hubei Han Chinese. Meta-analyses indicated that rs320 GG+TG genotype reduces and rs285 CC genotype increases the risk of MS.
