【Objective】To explore the genotype distribution of lipoprotein lipase polymorphism at Ser447→stop locus and possible association of them and dietary factors with the indicators of metabolic syndrome.【Methods】Physical examination and investigation by a 24h diet questionnaire were taken to permanent residents of a certain community in Shanghai.Resident's blood glucose and lipid levels were measured.The MS group was chosen by the results of the examination,and matched with control group by age and gender,the dietary intakes were compared and the possible relationship between dietary factors and metabolic syndrome was explored.The polymorphism of lipoprotein lipase gene at Ser447→stop locus was detected by PCR-RFLP method.The possible relationship between the polymorphism and the indicators of metabolic syndrome was explored. The possible association of dietary factors with the indicators of metabolic syndrome in different genotypes groups by adjusting age,gender and BMI were analyzed.【Results】①The men's blood pressure level,serum TG and TG/HDL-C ratio were significantly higher than women's,while serum LDL-C and HDL-C level were significantly lower than that of women.Men took more energy,protein and carbohydrate than women,and men had a higher intake of saturated fatty acids, mono-unsaturated fatty acids and cholesterol,as well as a higher proportion of animal fat.Men took more K and Vitamins,but less Na compared with women.②There was no significant difference in frequencies of genotypes or allele between the MS and the control,and between male and female.The genotype fiequencies of Ser447Stop SS, SX and XX were 85.6%,13.3%and 1.1%respectively,which were in agreement with Hardy-Weinberg equilibrium,and the allele frequency of Ser447 was 92.3%,stop447 was 7.7%.③The results of dietary investigation showed that the MS population took little dietary fibers,much Na,and was lack of Ca,K,vitamin B1 vitamin B2 and vitamin C.The energy intake came more from fats,and less from carbohydrates and protein.Less energy came from breakfast.The dietary structure of the ones with MS was unreasonable and should be adjusted.④After adjusting age,gender and BMI,the results of dietary investigation showed significant difference in protein and carbohydrate intakes between the people of different genotypes,the association between dietary intakes and blood glucose or lipid levels was significant in partial correlation regression,suggesting the possible interaction between the effects of the polymorphism of lipoprotein lipase gene at Ser447→stop locus and dietary factors with metabolic syndrome.【Conclusion】There might be a problem in dietary structure of the MS population. The energy proportion of three meals in a day and three nutrients which provide energy should be adjusted on the base of total energy controlling.The mutation of LPL gene Ser447→stop might be a protective factor for serum TG levels,and the people with stop447 allele genotype might tend to intake more protein and carbohydrates.The interaction between the effects of the polymorphism of lipoprotein lipase gene at Ser447→stop locus and dietary factors with metabolic syndrome might exist actually.A personalized intervention program should be necessary in the control of MS because of dietary predisposition of gene polymorphism.
