Objective:Positive promotion effects of LMHFV(Low-magnitude high-frequency vibration) have been proved for fracture healing in the fields of cytology and biology. However, there is few research about how LMHFV effects the fracture injury of live big size mammal.Besides, the lack of articles' talking about the comparisons between the different vibration parameters and the difference between internal fixation and external fixation in the present literatures really evoke my interests doing the first initial research. My research is trying to explore the appropriate motion parameter of LMHFV when using it to facilitate the fracture healing process of long bone, and provide trustful theoretical evidence for clinical experiments through comparing the healing results of fracture healing's different managements.Methods:The samples were comprised of 12 healthy female goals with same age as 8 months and their weights ranged from 17 kg to 20 kg. After equally distributing all these samples into six different groups at random,Metacarpal and metatarsal transverse osteotomy had been implemented to them under same anesthetic and aseptic condition. I used my grouping process according to different managements as following:Group A:external fixation control group;Group B: internal fixation control group;Group C:external fixation and vibration group a;Group D:external fixation and vibration group b;Group E:internal fixation and vibration group a;Group F:internal fixation and vibration group b.Vibration with certain parameters and form had been implemented to each vibration group one week later of the operations.And all of the samples were put on the custom stent to make the limbs off the ground.X-ray pictures combined with serum samples had been taken before the surgeries and 1 day, 2 weeks, 4 weeks, 6 weeks, and 8 weeks after the operations.Air embolism method had been used for killing all these samples at the end of 8th week of the operations after picking out the serum of each goat for observing the osteogenesis markers and assessing the mechanical property of the samples' bone with mechanical instrument.Results:(1)The whole samples were observed no obvious displacement of the segment on X-ray pictures.And the wounds healing were first intention,without infection.(2)Imaging dynamic observation:No obvious injury or skeletal deformity were found on X-ray pictures of the samples before they underwent operations;It showed on X-ray pictures that the calluses of the vibration groups were far more than that of the control groups since the former accepted vibration at every point-in-time of observation,further more,the calluses of the Group C?E were found more than that of Group D?F;At the end of the experiment,there is not any obvious differences between Group A and Group B,while the samples of the two groups both acquired more calluses which can be telled easily than they did at the begaining;Marrow cavity recanalization was found on some of the samples' X-ray pictures of the Group C?E,while it didn't showed on the pictures of the rest samples.(3)Dynamic observation of the osteogenesis markers in serum:Two weeks after the operations,the concentration of serum osteocalcin of the vibration groups were observed higher than the control groups,while there was not any obvious difference among the four vibration groups,on the contrary the concentration of serum TRAP5 b of vibration groups were lower than that of control groups;Four weeks after the operations,the relationship among all groups on the concentration of serum osteocalcin are as follows:Group A?B>C>E>D>F,as to the concentration of serum TRAP5b:Group A>B>D>F>C>E;Six weeks after the operations,the relationship among all groups on the concentration of serum osteocalcin:Group C ? E>F>D>B>A,as to the concentration of serum TRAP5b:Group A>B>D>F>C>E;Eight weeks after the operations,the relationship among all groups on the concentration of serum osteocalcin:Group C>E>F>D>B>A,as to the concentration of serum TRAP5b:Group E>C>F>D>B ? A.(4)The appearance of each samples' callus:Dissect all samples at the end of the experiment,cut out the injured limbs,bigger and harder,more smooth calluses were found on the surface of the samples' limbs which accepted vibration.By contrast,the calluses of control groups' sample were more fragile with lots of pores.(5)Mechanical property test:It showed that the modulus of elasticity of the vibration groups' metacarpal and metatarsal bone on the fracture end were a little larger than that of control groups' bone when tested them with mechanics apparatus(p>0.05),while the modulus of elasticity of the vibration groups' metacarpal and metatarsal bone which cut from 10 mm and 20 mm away from the fracture end were much larger than that of control groups' bone(p<0.05).meanwhile,there was not any obvious difference among the four vibration groups(p>0.05).Conclusion:1.Both two motion parameters had increased the healing process of fractures in the samples,and the parameter a had better outcomes than parameter b. Besides, few differences had been found between the two fracture stabilizing managements.2.The dynamic monitoring serum showed that the concentration of serum osteocalcin was higher of the goats in the vibration group than the control group at the beginning and ending periods, and lower in the metaphase. This result states that more effective management stimulated osteoblasts' proliferation and activation for the fracture healing process of the vibration groups' samples during the recovery phase consistently; another result demonstrated that the concentration of the serum TRAP5 b of the experimental group was lower than the control group at both the opening phase and the metaphase but higher at the ending phase, which hints that these samples that had been stimulated by LMHFV suppressed the osteoclasts' activity for accelerating the speed of osteogenesis at the opening phase.Stimulated at the opening phase,at the next phases, the experimental group samples were faster for experiencing fracture recovery.3. The calluses of the vibration groups' goats were more compact, structured, and smooth than the control group after certain healing time, which proves the porosis under stimulation is more effective for the vibration group.4. There is no obvious difference between vibration groups and control groups on the modulus of elasticity on the fracture end,but the differences increased with the distance away from the fracture end.Nevertheless, conclusion is hard to tell that there is any difference for the samples with different stabilizing management under the same motion parameter.Above all,LMHFV with proper parameters(as parameter a in our experement) can accelerate the speed of osteogenesis on stable long bones' fracture while healing,while there is not any obvious difference among the samples with different stabilizing management under the same vibration parameter.
