| Objective:To explore the relationship of atherosclerosis and micro RNA(miRNA)in hypertensive patients with carotid artery atherosclerosis(CAS),understanding Predictive Value of miRNA in the early diagnosis for atherosclerosis.Methods: Total 400 hypertensive patients were selected and divided two groups based on carotid artery atherosclerosis: hypertension group and hypertension with carotid artery atherosclerosis group according to blood pressure measurement and ultrasound carotid artery intima-media thickness(cIMT).Each group contained 200 cases.Peripheral blood was collected and plasma was separated.Total cholesterol(TC),triglyceride(TG),high density lipid-cholesterol(HDL-C)and C-reactive protein(CRP)were determined by automatic biochemical analyzer,and interleukin-6(IL-6)by enzyme-linked immunosorbent assay(ELISA).Extraction of total plasma miRNA,the expression differences of hsa-mi R-33,hsa-miR-125a-5p,hsa-miR-126,hsa-miR-145 and hsa-miR-146 a were detected with real-time qPCR assay.The hypertensive patients without carotid artery atherosclerosis were follow-up.The plasma lipid,CRP,IL-6 and miRNA were analyzed in the newly patients occurred carotid artery atherosclerosis.The relationship between the changes in these parameters and carotid artery atherosclerosis were analyzed.Results: HDL-C in the hypertensive patients with carotid artery atherosclerosis group were significantly lower than hypertensive patients without carotid artery atherosclerosis group(all P < 0.05).The selected 5 miRNAs relative expression levels between the two groups also had significant differences(all P < 0.05).The up-regulated expression of miRNA were hsa-miR-33 and hsa-mi R-146 a,and down-regulated expression mi RNAs were hsa-mir-125a-5p,hsa-mi R-126 and hsa-miR-145 in the hypertensive patients with carotid artery atherosclerosis group.In the follow-up of patients,plasma HDL-C,CRP and IL-6 have significant differences between patients with and without newly occurred atherosclerosis and plaque(all P < 0.05),and between these parameters and carotid artery atherosclerosis showed positive correlation.5 mi RNAs relative expression level also have significant differences between Two groups(all P < 0.05).The has-miR-33 and has-miR-146 a and carotid artery atherosclerosis and plaque formation showed positive correlation,has-miR-125a-5p,has-miR-126 and has-miR-145 showed negative correlation.Conclusion: In hypertensive patients with carotid artery atherosclerosis,HDL-C,CRP and IL-6 can be used as a serum marker,and hsa-miR-33,hsa-miR-125a-5p,hsa-miR-126,hsa-miR-145,hsa-mi R-146 a relative expression differences can be used as a marker in the early diagnosis of atherosclerosis. |
