Objective: To optimize the best extraction and purification technology of total flavonoids from Aster souliei Franch; To study mechanisms of acute mountain sickness and provide experimental evidence for clinical application.Method: In our Preliminary research results, effective part of Aster souliei Franch is total flavonoids which could prevente and cure of acute mountain sickness. Extraction technology conditions of total flavones was optimized by orthogonal test. To optimize the best purification technology of macroporous adsorption resin,normbaric hypoxia experimental model were used to explore anti-hypoxia effect and best effective dose,mice is induced acute mountain sickness by Acute decompression Hypoxia experiments, By measuring the hypoxic injury and antioxidant biochemical indices,To study effect which is prevent and cure acute mountain sickness of the total flavonoids from Aster souliei Franch,To analyze Mechanism of total flavonoids from Aster souliei Franch by west blot.Result:1. The optimum extraction conditions were as follows: using Twelve Times the amount of water and heated to reflux for 2 times, 2 hour for each time. The ratio of extraction can reach 24.58%, total flavonoid content can reach 29.7%.2. Static adsorption capacity and Dynamic adsorption capacity were employed as examine items, The LX-60 macroporous adsorption resin showed the best desorption rate and was chosen as the material for the purification of total flavones. The optimum purification conditions were as follows: sample liquid concentration is 4.124 mg /mL,the sample flow rate of 1.0 ml/min, After standing for 15 min, 2BV with distilled water, 60% ethanol 4BV according to 1.0 ml/min flow rate to elution, by purity indicators to determine the diameter and height ratio2.5: 8.3. In normbaric hypoxia experimental,Compared to the model group,three dose of total flavonoids from Aster souliei Franch enhanced the survival time of mice and in dose-dependent manner,mice of high dose group can enhanced 32% and the optimum dose is 500mg/kg.4. In Acute decompression Hypoxia experiments,Compared to the normal group,Hypoxia could increased the content of MDA,LDH,H2O2 and CK in heart and brain. Compared to the model group,total flavonoids from Aster souliei Franch could decrease the content of MDA,LDH,H2O2 and CK.5. In Acute decompression Hypoxia experiments,Compared to the normal group,Hypoxia could decrease the activity of CAT,SOD,GSH and GSH-PX in heart and brain. Compared to the model group,total flavonoids from Aster souliei Franch could increased the activity of CAT,SOD,GSH and GSH-PX.6. In Acute decompression Hypoxia experiments,Compared to the normal group,Hypoxia could increased the content of Nrf2?HIF-1??VEGF and caspase 3, decrease the content of bcl 2/bax and SOD in brain;Compared to the model group,total flavonoids from Aster souliei Franch could decrease the content of HIF-1??VEGF and caspase 3,increased the content of Nrf2?SOD and bcl 2/bax.Conclusion:1. Using the preferred extraction process, It can greatly improve the extraction rate of total flavonoids, the process is simple, feasible and provide drug source for pharmacological experiments.2. To study of Purification process, It developed a precise and effective purification methods, this process can be used for industrial production of total flavonoids from Aster souliei Franch.3. normbaric hypoxia experimental show closed total flavonoids from Aster souliei Franch can effectively control anoxic damage in whole, this experiment provide a reliable premise for pharmacodynamic research4. Acute decompression Hypoxia experiments indicate total flavonoids from Aster souliei Franch may reduce lipid peroxidation, anaerobic glycolysis and oxygen free radicals by hypoxia induce.5. Acute decompression Hypoxia experiments indicate total flavonoids from Aster souliei Franch can improve the activity of antioxidant enzyme, increase the body's antioxidant capacity and resist oxidative stress.6. It confirmed total flavonoids from Aster souliei Franch have a role and influence in antioxidant pathway, anti-hypoxia pathway and inhibition of apoptosis pathways for protein determination in three areas.
