| Fruit and vegetables are indispensable in people’s daily life.In recent years,with the increasing improvement of living standards,the physiological and biochemical functions of the active ingredient in fruit and vegetables,such as prevention effects on some chronic disease,have attracted a great interest and the research of fruit and vegetables health food has become one of the hotspots of the current food industry development.Currently,there is few report on the relationship between common fruits and vegetables and blood uric acid system,and there is also few health food product in lowering uric acid levels made from fruit and vegetables sources.Therefore,it is important and significant to investigate the activity of fruit and vegetables,and to determine the main active ingredient.The contents are as follows:In this paper the inhibitory effect of 70%ethanol extracts of 40 kinds of common vegetables and 22 kinds of common fruit on xanthine oxidase activity in vitro were studied.At the dose of 500 mg/L,39 kinds of extracts exhibited inhibitory activity effects on xanthine oxidase.Five kinds of the fruit and vegetables ethanol extracts with stronger activities among all 62 fruit and vegetables’ extracts were that of maybush,Artemisia Selengensis Turxcz,caraway,pimiento and snow peas,repectively,and their correspongding inhibiting rates were 69.8±0.16%,65.7±0.1%,44.2±0.08%,40.1±0.16%and 36.6±0.1%,respectively.Then the IC50,were 251.4 mg/L,330.8 mg/L,599.7 mg/L,613.2 mg/L and 646.9 mg/L,respectively.Then enzyme activity of these five different concentration of fruit and vegetables alcohol extract in different substrates(xanthine)concentration were determined.According to the Lineweaver-Burks double reciprocal equation plotting,the kinetics of enzyme catalytic reaction accounted for the changes of michaelis constant arguments(Km)and the maximum reaction rate(Vm)to determine XOD inhibitory type of the Artemisia Selengensis Turxcz,caraway and snow peas alcohol extract.For all competitive inhibition,the inhibition constants Ki were 39.7 mg/L,71.2 mg/L and 73.3 mg/L.XOD inhibitory type of maybush,snow peas alcohol extract were determined as hybrid inhibition and their inhibition constants Kj were 32.4 mg/L and 76.8 mg/L,and Kis were 511.5 mg/L,and 454.5 mg/L.Inhibitor were mainly combinated on the enzyme while a small part of may be combinated on the complex of enzyme and substrate.Meanwhile,the total content of flavonoids in 62 kinds of fruit and vegetables ethanol extracts were determined respectively.Results showed that there were flavonoids in 62 kinds of fresh fruit and vegetables,flavonoid content ranged 31.1 ±0.14~382.8±0.28mg/g,15 kinds of samples among 62 kinds of fruit and vegetables ethanol extracts were detected as more than 100 mg per gram fresh content,accounting for 24.2%,5 kinds of samples were detected as more than 200mg,which were Artemisia Selengensis Turxcz,maybush,caraway,pimiento and snow peas.And the flavonoid content in Artemisia Selengensis Turxcz were the highest.It is speculated that flavonoids in the fruit and vegetables extracts may be the main inhibitory substances of xanthine oxidase.By established mice model of high uric acid hematic disease,high uric acid hematic disease preventing activity in mice of five kinds of crude extracts were estimated.In vivo animal experiments results showed that all the five kinds of extracts had uric acid reduction effect on high uric acid hematic disease model mice which caused by oxygen oxazine acid potassium,and also could inhibit the activity of liver xanthine oxidase.The extract of Artemisia Selengensis Turxcz had the most remarkable effect.Blood uric acid levels and xanthine oxidase activity of Artemisia Selengensis Turxcz alcohol extract of high dose(blood uric acid 10.72±0.85mg/L,enzyme activity 44.58±0.96U/g prot)were the closest to allopurinol group(blood uric acid 3.78±0.72mg/L,enzyme activity 40.31±0.79U/g prot).The main components of analyzing Artemisia Selengensis Turxcz alcohol extract were analyzed by the LC-MS technology,which were speculated as chlorogenic acid derivatives and flavonoids substances such as rutin. |
