Object: This study to compare the connexin expression differences of renal interlobar artery ofdifferent weeks of spontaneously hypertensive rats. To investigate connexin changes inhypertension renal artery.Methods: Twenty-four SHR were randomly divided into three groups(4weeks,12weeks,48weeks),and24sex-and age-matched Wistar rats were assigned to control groups.Tail arterialSystolic Blood Pressure(SBP) was measured by BP-6noninvasive determination of blood pressuremonitor;Pressure myograph system was used to observe the vasoconstriction induced by KCl andPE (endothelial independent vasoconstrictor), and the changing of vasoconstriction after applying18-?GA (gap junction blocker) in acute separated renal interlobar artery of SHR and Wistar rat,Protein expression of Cx40and Cx45was detected by Western-blot.Results:(1) Contraction reaction induced by KCl and PE were in a concentration-dependent wayin renal interlobar artery of SHR and Wistar rat. The contraction reaction induced by KCl and PEwere higher in SHR than Wistar rats respectively (P<0.05), there were significant difference in12weeks(P<0.05). Compare with4weeks, the initial concentration and the maximum reactionconcentration of KCl and PE were move up at12and48weeks(P<0.05).(2) After applicated with18?-GA (100?mol/L), the contraction reaction induced by KCl and PEwere suppressed in SHR and Wistar rats(P<0.05), the inhibit amplitude were higher in SHR thanWistar rats respectively (P<0.05), and more significantly in12weeks (P<0.05).(3) Western-blot detection: Cx40expressions were no difference between different weeks ofWistar rat(P>0.05), SHR decreased with increase of age, both4and12weeks SHR were increasedand48weeks old was decreased compare with age-matched Wistar rats(P<0.05). The Cx45expression was first increased and then decrease, SHR expressed higher than Wistar rat(P<0.05)?and more significantly in12weeks (P<0.05).Conclusions:(1) The contraction reaction was higher in SHR than Wistar rat; it may be associatedwith enhanced gap junction communication in renal interlobar artery of SHR. Hypertension andage can be affected the function and expression of gap junction of rat renal artery.
