Objective Observation of Runx3, TGF-β1and CD83(the marker for completely matureDCs)in patients with irritable bowel syndrome (IBS group), patients with ulcerative colitis(UC group) and the normal volunteers (control group) the expression in sigmoid colon tissuesMeanwhile,exploring the Runx3、TGF-β1and dendritic cells the relationship between thethree of them in IBS and UC and providing clues to the pathogenesis of IBS and UC.Methods1、Real-time fluorescence quantitative PCR in40cases of irritable bowel syndromepatients,40cases of ulcerative colitis patients and40cases healthy volunteers as controlgroup to detecte the expression level Runx3mRNA, TGF-β1mRNA and CD83mRNA insigmoid colon mucosal and to analyze the relationship between the three of them in IBS andUC.2、Immunohistochemistry technique in40cases of irritable bowel syndrome patients and40cases normal volunteers the protein expression level Runx3, TGF-β1and CD83in sigmoi-d colon mucosal.Result1、Compared with the control group, There was no significant difference between theexpression of Runx3mRNA and TGF-β1mRNA (P>0.05) in irritable bowel syndromepatients. The expression level of CD83mRNA was higher in patients with irritable bowelsyndrome patients than in control group (p<0.05) and were associated negatively with Runx3mRNA levels (r=-0.361, p<0.05). Meanwhile, Runx3mRNA level was associated positively with TGF-β1mRNA expressions in irritable bowel syndrome patients (r=0.402, p<0.05).However, there is no correlation between the mRNA expression of TGF-β1and CD83(p>0.05).2、Compared with the control group, the expression level of Runx3mRNA、TGF-β1mRNA in patients with ulcerative colitis were lower than in control groups (P<0.05), and theexpression level of CD83mRNA in patients with ulcerative colitis were higher than in heal-thy controls (P<0.05);The expressions level of Runx3mRNA and TGF-β1mRNA waspositively correlated in ulcerative colitis patients(r=0.629, P<0.05);The expressions level ofRunx3mRNA and CD83m RNA was negatively correlated in the ulcerative colitis patients(r=-0.456,P<0.05);The expression level of TGF-β1mRNA and CD83mRNA was negat-ively correlated in ulcerative colitis patients(r=-0.364, P<0.05).Conclusions1、Runx3and TGF-β1needs to be further studies in irritable bowel syndrome patients;Theincrease of abnormal dendritic cells might influence the occurrence and development ofirritable bowel syndrome patients; TGF-β1signal pathway might not be involved in Runx3-regulated maturation of dendritic cells in irritable bowel syndrome patients;2、Runx3、TGF-β1and dendritic cells are closely related with the ulcerative colitis;Runx3gene may regulate TGF-β1-mediated dendritic cells function in ulcerative colitis patients.
