Aims:To research the expression of RUNX3 and COX-2 in normal colonic mucosa(NCE),colon adenoma(CAP),and colon cancer(CC)tissues.To analyze the relationship between RUNX3 and COX-2 and colon cancer-related clinicopathological data,and to explore whether the expression of the two proteins in cancer tissues is related,in order to provide new ideas for future research on the pathogenesis and targeted therapy of colon cancer.Methods:1.Collecting 40 cases of colon adenomatous polyp(CAP)diagnosed by the Pathological Diagnosis Center of the chenzhou first people’s hospital between January 2019 to June 2020 and 46 pairs of colon cancer(CC)and normal colonic mucosa(NCE)tissue chips produced by Shanghai Tufei Biotechnology Company were used as the research objects.2.The immunohistochemical method was used to detect the expression of RUNX3 and COX-2 in the three groups of colon mucosal tissues.The data of this experiment was statistically analyzed by SPSS19.0 statistical software.Results:1.The expression of RUNX3 in CC group was low,the positive expression rate of RUNX3 in NCE group,CAP group,and CC group was 89.1%(41/46),62.5%(19/40),32.6%(15/46),and there was statistical difference among the three groups(P<0.05);the expression intensity of RUNX3 in the three groups was also statistically different(P<0.05),the expression intensity of RUNX3 in the CC group was lower than that in the CAP group(P<0.05)and the NCE group(P<0.05),and the expression intensity of RUNX3 in the CAP group was lower than that in the NCE group(P<0.05).2.The expression of COX-2 in CC group was high,the positive expression rate of COX-2 in NCE group,CAP group,and CC group was15.2%(7/46),45.0%(18/40),76.1%(35/46),and there was statistical difference among the three groups(P<0.05);the expression intensity of COX-2 in the three groups was also statistically different(P<0.05),the expression intensity of COX-2 in the CC group was higher than that in the CAP group(P<0.05)and the NCE group(P<0.05),and the expression intensity of COX-2 in the CAP group was higher than that in the NCE group(P<0.05).3.The expression of RUNX3 in the CC group was not significantly correlated with the patients’ gender,age,tumor size,and degree of differentiation(P>0.05),but it was correlated with the depth of tumor invasion,lymph node metastasis and clinical TNM stage(P<0.05).4.The expression of COX-2 in the CC group was not significantly correlated with the patients’ gender,age,tumor size,and degree of differentiation(P>0.05),but it was correlated with the depth of tumor invasion,lymph node metastasis and clinical TNM stage(P<0.05).5.The expression of RUNX3 and COX-2 in the CC group was negatively correlated(r=-0.384,P<0.05).Conclusion:1.The expression of RUNX3 in CC group was low,and the expression of RUNX3 in normal colon mucosa,colon adenoma,colon cancer showed a downward trend.2.The expression of COX-2 in CC group was high,and the expression of COX-2 in normal colon mucosa,colon adenoma,colon cancer showed an upward trend.3.In colon cancer,the low expression of RUNX3 and the high expression of COX-2 are closely related to the depth of tumor invasion,lymph node metastasis and clinical TNM stage.4.The expression of RUNX3 and COX-2 in colon cancer is negatively correlated,and the combined detection of the two may help the diagnosis,condition judgment and targeted therapy of colon cancer.
