Clinical Observation Of Qingchinghuashi United Mesalazine Treating Ulcerative Colitis And Mechanism Research Of The Interleukin-23/17Axis

Objective The aim of this study is to observe the clinical therapeutic effect of QingChingHuaShi Method in the treatment of ulcerative colitis; determinate the expression of IL-23and IL-17in colonic tissue and plasma of the patients, and to explore the possible action mechanism of UC.Methods Forty patients who were diagnosed as in active stage of UC by colonoscopy and diagnosed as the syndrome of internal stasis of dampness and heat by syndrome differentiation of traditional Chinese medicine were divided into two groups.In the control group,20patients were treated with Mesalazine SR Granules, while the other20patients in the treatment group were treated with QingChingHuaShi Method besides Mesalazine SR Granules. The course of treatment is3months. When the treatment is over, compare the clinical curative effect?the states of the intestinal mucous membrane and the remission rate. Then test the expression of IL-23and IL-17in colonic tissue of the patients by the Immunohistochemistry method, meanwhile test the expression of IL-23and IL-17in plasma of the patients by Enzyme Linked ImmunoSorbent Assay.Results The treatment group and the control group can improve the main symptoms of individual(diarrhea, mucous bloody stool, abdominal pain), clinical symptom total score, the comprehensive curative effect, TCM syndrome, P values were less than0.05, and the effect of the treatment group is better than the control group's, P values were less than0.05; the treatment group colonoscopy and pathological efficacy was superior to the control group (P<0.05), and the remission rate of a group was higher (P<0.05),too. Two treatment methods can reduce expression of IL-23and IL-17in colonic1tissue of UC patients by immunohistochemical method, but the effect of the treatment group is better than the control group's (P<0.05); ELISA method to detect two kinds of methods of treatment reveal that two therapies also can effectively reduce the concentration of IL-23and IL-17in plasma of UC patients, but the treatment group compared with the control group (P<0.05) has advantage.Conclusion The curative effect of QingChingHuaShi method combined with Mesalazine SR Granules treating ulcerative colitis patients is better than that of Mesalazine SR Granules only.Role of the mechanism may be related to the inhibition of proinflammatory cytokines IL-23and IL-17expression in plasma and colon tissue of UC patients, suggesting our QingChingHuaShi Method may play a role in the effect of IL-23/IL-17axis.
