Poria cocos belongs to the genus Eumycophyta,Basidiomycotina,Hymenomycetes,Aphyllophorales,Polyporaceae,Wolfiporia.Also known as Fu Ling,pine Bacon,Songbaiyu,Fu Tu,etc.Depending on different production area,it also named as An Ling,Yun Ling,Chuan Ling,Min Ling and so on.For a long time,Poria cocos cultivation strains are obtained by the separation of sclerotium tissue so that caused the quality of Poria cocons were variant;the introduction of each Poria production areas was frequent,which lead to "the same name but the different strain,the different name but the same strain",the problem may result the quality and cultivation in hidden dangers.So far,the domestic Poria cocos germplasm resources and its genetic diversity has not been systematically studied that brought difficulties to resources utilization and the breeding of fine strains.In this paper,we collected 10 kinds of Poria cocos strains and investigated the biological characters,such as rejuvenation,growth rate,sclerotia yield agronomic traits,sclerotia water-soluble polysaccharides and total triterpenoids,and studied on the genetic polymorphism of the tested strains by ISSR.After the rejuvenation of the strain,the strain had a large difference before and after the rejuvenation,and the overall performance was stronger than that before the rejuvenation,the density of the hyphae before the rejuvenation,the climbing ability and the stout of the aerial mycelium.In this paper,ten biological strains of rejuvenation,growth rate,sclerotia yield agronomic traits,sclerotia water-soluble polysaccharides and total triterpenoids were investigated,and the ISSR molecular markers The genetic polymorphism of the tested strains was studied.After the rejuvenation of the strain,the strain had a large difference before and after the rejuvenation,and the overall performance was stronger than that before the rejuvenation,the density of the hyphae before the rejuvenation,the climbing ability and the stout of the aerial mycelium.When the strain of Poria cocos was grown on the medium,the mycelium density,the degree of development of the bacteria and the degree of development of the aerial mycelium were different.The growth rate of the same strain on different media was different.In general,the growth rate of Poria cocos was higher than that of the medium in maltose medium.The growth rate of the tested strains on the common medium was between 10.35±0.07 days and 15.58±0.11 days before rejuvenation.The growth rate of the medium on the common medium was 6.15±0.05-8.31±0.05 days.The results showed that the average weight of the bacteria was 1.142±0.03 kg,and the yield was higher than that of other strains.The contents of water-soluble polysaccharides in the tested strains were between 3.57 mg / g and 5.83 mg / g,and the total triterpenes were between 8.32 mg / g and 11.08 mg / g.Variance analysis showed that these biological traits showed significant or very significant differences.After 10 pairs of Poria strains between the two pairs of antagonistic test,obtained six groups produced significant antagonism,which were 86 and Yunnan Poria,Hunan 28 and Fu 1,5.78 * and Fu Tai 86 and with 578 And Fu 1,Hunan 28 and Fu 2The primers were designed according to the information of Poria genome and transcriptome,and 5 pairs of polymorphic primers were screened.According to the polymorphic bands,all the 10 tested strains were clustered,which could be divided into six categories.The results showed that the germplasm resources of Fulu and other germplasm resources showed great difference in biological traits,which provided the material and basis for the breeding of Poria cocos,but the genetic background of Poria germplasm resources was extremely narrow,genetic diversity is not rich,need to strengthen the collection and protection of Poria germplasm resources.
