Poria cocos(Schw.)Wolf is a widely used traditional Chinese medicine with large demand.The triterpenoids are important metabolites of the mushroom,which are also important indicators for the quality control of the medical material.In this study,we tried to study the effect of periodic fluctuation of light and temperature,as well as adding different kinds of vegetable oil,on the accumulation of tritepenoids.The main results of this study are summarized as follows:(1)To explore the effect of stimulation of light and temperature on the biomass and triterpenoids content,fermented conditions were conducted with different patterns of periodic low/high temperature,dark/bright light treatment and the combination of these two.The results exhibited that the combination of light and temperature treatment increased biomass up to(24.78%)and triterpenoids(34.61%)compared to the control group.(2)After adding 1% peanut oil,rapeseed oil,palm oil,soybean oil,olive oil and coix seed oil,the biomass of alfalfa and the total amount of triterpeniods were significantly increased.The best effect was achieved with olive oil.The biomass and total triterpeniods content were 2.20 times and 2.29 times higher than that of the control,respectively.(3)Since the olive oil showed the best effect,we further optimized the culture condition in the presence of olive oil.It showed that the best inoculation time is at the beginning of a culture with a dosage of 2%,inoculum rate 8% for a fermentation time of 8days.The biomass and total triterpenoids content were 4.15 times and 4.09 times than that of control,respectively.
