Preliminary Study On Pharmacological Action Of Compound Fructus Cnidii Lotions On Fungal Skin Disease

The fungal skin disease of pet was one of the common diseases in pet clinic and it's mainly caused by superficial fungal.With the widely application of anti-fungi drugs recently,it became more and more difficult to treat the fungal infections for the increasing drug-resistance.Chinese Pharmacology studies have shown that there are significant effects of antibacterial and antifungal in many antipyretic and drugs for external application,meanwhile,the drugs are also characterized with broad antimicrobial spectrum and low drug-resistance.Our proprietary formula are composed of cnidium,Fructus Kochiae,Stemona japonica,Garden Burnet,Fructus Toosendan,Polygonum,coptis chinensis,etc.It turned out that the proprietary formula had positive treatment effect on fungal skin diseases in pet,while its inhibitory mechanism was still not clear.The compound prescription of cnidium lotion and osthole are the tested drugs in my study and terbinafine and caspofungin commonly used in clinic were the controlled groups.The study aims at evaluating the pharmacological action and safty of compound prescription of cnidium lotion through the tests including the minimum inhibitory concentration measurement,the effect of cell membrane and cell wall study of microsporumCanis,mouse ear swelling test and mouse writhing test,local skin acute toxicity test,local irritation and allergic test.Result as follows:(1)The result of antibacterial test in vitro indicated that microsporumCanis directly enter the stage of exhaustion without entering normal growth cycle on minimum inhibitory concentration of compound prescription of cnidium lotion;the number of live bacteria declined steeply during 12-24 h and all the live bacteria were dead during 24 h under drug effects.(2)The result of the effect of cell membrane and cell wall: the study showed that the content of ergosterol in compound prescription of cnidium lotion group and terbinafine group were great significantly lower than that in osthole group and control group(P<0.01),and the content of ergosterol in compound prescription of cnidium lotion group was great significantly lower than that in terbinafine group(P<0.01);the content of 1,3-?-d-glucan and chitin in compound prescription of cnidium lotion group,osthole group and caspofungin group were great significantly lower than that control group(P<0.01).(3)The result of anti-inflammatory and analgesic test showed that the ear swelling and swelling rate of mice in compound prescription of cnidium lotion group were significantly declined(P<0.01);the twisting time in high dose,medium dose of compound prescription of cnidium lotion group and western medicine control group(Diclofenac Diethylamine Emulgel)were significantly delayed(P<0.01)compared with the control group,the times of writhing were declined significantly(P<0.01).Compared with western medicine control group(Diclofenac Diethylamine Emulgel),writhing occurred in medium and low dose group of compound prescription of cnidium lotion was significantly earlier(P<0.01).(4)The result of skin safty study showed that there was no die of guinea pig at 1 h,24 h,48 h,72 h and 7 d after removing drug in complete skin group and damaged skin group,and there was no abnormal phenomena and erythema,edema at smear site,indicated that compound prescription of cnidium lotion had no acute skin toxicity;there was no edema,hemorrhage,rough skin in complete skin group and damaged skin group after continuous delivery 1 week,the mean value of skin irritation was 0.30,indicated that compound prescription of cnidium lotion had no skin irritation;the result of allergy test showed that there was no erythema,edema,and other skin allergy in compound prescription of cnidium lotion group.Conclusion:(1)Compound prescription of cnidium lotion had better antibacterial effect on microsporumCanis and its inhibitory mechanism may include inhibiting inherent substance of the cell membrane and cell wall of microsporumCanis.(2)Compound prescription of cnidium lotion had good anti-inflammatory and analgesic effects.(3)Compound prescription of cnidium lotion had no acute toxicity,irritation or allergic reactions,it was safety in clinical.
