Until this resport, researches on toxicity of Adprin lacked a relative systematic one in our country. In order to evaluate the safety of Adprin, some systematic toxicological experiments were carried out by the principles and methods of toxicology which were based on Adprin’s physico-chemical properties and clinical characteristics.In the test, the Kun Ming mice and hyline chicken were taken as experiment animals and the toxicity of Adprin was studied by intragastric administration. Firstly the LD50for Adpin was determined in the acute toxic test and then the30d accumulative toxicity test, micronucleus test, sperm abnormality test were performed on mice. Acute toxic and subacute toxic test of Adprin were also carried out on chicken, the target animal.Acute toxic test results showed that:median lethal dose (LD50) of Adprin in mice is43.76mg/kg and its95%confidence interval range is35.73~53.58mg/kg. According to the WHO recommenged classification of acute toxicity on exogenous chemical compounds (5level) it indicated that Adprin is high-poisonous substance on mice.Accumulation of toxicity test results showed that:Storage coefficient of Adprin in mice is3.76, the accumulative dose is164.54mg/kg which revealed a modereate capacity of accumulative toxicity on mice according to accumulative grading standards. And accumulation toxicity caused mice growth inhibition.PCE microcleus test in Bone Marrow cells on mice is negative and there is no significant defference compared with negative control. So Adprin is no chromosome clastogen and no spindle toxic effect on mice.Sperm abnormality test on the male mice reproduction cells is negative which showed that Adprin was non-mutagenic activity on germ cell.Acute toxicity test results showed that:median lethal dose(LDso) of Adprin in hyline chicken is108.44mg/kg and its95%confidence interval range is95.17~123.56mg/kg. According to the WHO recommended classification of acute toxicity on exogenous chemical compounds (5level) it indicated that Adprin is mid-poisonous substance on chicken. The liver may be the major target organ of acute toxitity on chicken.Subactute toxicity test results showed that mice has non-subactue toxicity with Adprin at dose of1/80~1/20OLD50for30d administration. And Adprin has no effect on chicken growth and immunological function. The function of liver and kidney was normal. These experimental results demonstrates that we need to restrict the dose of Adprin and comply with the withdrawal period in the clinic. And stopping the drug before slaughter.
