| Phellodendron amurense Rupr.Belongs to Rutaceae Branch Berberis deciduous trees,alias P.amurense,it is Seppi medicine,is commonly used in the main source of Chinese medicine cork.P.amurense is one of the relict plants of the Tertiary tropics.It has very important research and academic value and is currently listed as a national secondary protected plant[1].P.amurense is also a good nectar plant,its flowers can provide excellent honey for the bees,is widely used in medicine and industry[2].In the endangered state,P.amurense has been listed as China’s rare plant,but also has its own resources.Plants is the national level to protect wild herbs species.Because P.amurense is both a rare medicinal tree species and an important timber species is also an ancient remnant plant,and berberine medicinal,material and ornamental value in oneself,with a high protection and development and utilization of vision.Due to long-term deforestation,the current number has been very little.Therefore,the protection and research of P.amurense.has important scientific significance.In this paper,the seeds of P.amurense were studied by physiology and proteomics,the main results are as follows:1、P.amurense seed is a physiological dormant seed,through the low temperature stratification,it takes three months to gradually lift its dormancy.2、The seed dormancy was gradually broken with the increase of low temperature stratification time,and the soluble sugar in the seeds showed the trend of increasing first and then decreasing,and the content of starch and crude fat decreased,and the content of starch Acid content showed an upward trend.3、The activity of key enzymes in the process of lamination treatment also changed,SOD activity decreased,POD activity and CAT activity increased first and then decreased.Acid phosphate esterase showed a trend of ascending trend,SDH activity showed a decreasing trend,glucose-6-phosphate dehydrogenase showed a tendency to rise first and then decrease,and these changes were related to seed dormancy.4、Proteomics analysis of P.amurense seeds with different stratification treatments was carried out using pH4-7 IPG strips.A high-resolution and reproducible protein 2-DE map was obtained by a reasonable focusing procedure.Silver dye software analysis,each glue can detect more than 500 protein spots.The results of the analysis showed that 28 protein abundance differences were detected in all treatments and controls.The six proteins were identified by mass spectrometry,and the six proteins were searched by NCBI BLAST and referenced to the GO classification criteria and related literature.The function of the protein of P.amurense was explained in different low temperature stratification treatments classification.They were involved in amino acid metabolism(17%),storage protein mobilization(17%),starch metabolism(17%),glycolysis related(33%). |
