Study On The Mechanism And Effect Of Huoqi Perfusate For The Reproductive Function Of Female Mouse

The Chinese veterinary Huoqi perfusion was developed according to the principles of syndrome differentiation and treatment in Chinese Veterinary Medicine.And it,s made of 8 kinds of traditional Chinese medicine,such as Actinolite,Cuscute Senmen and Epimedii Foliumetc.The treatment principle ofthis medicine was nourishing liver and tonifiying kidney and aphrodisiac.The clinical test showed that,this medicine could effectively treat Stillness of the ovary and persistent corpus luteum in dairy cows.Then the toxicology research showed that the preparation is safe,and it has obtained the new veterinary medicine certificate.In order to study the mechanism of Huoqi Perfusate,the following experiments were carried out.1.Effect of the Huoqi Perfusate on Reproductive Function of female mouseTo explore the effects of the Huoqi Perfusate on reproductive organ growth,hormone and antioxidant levels in female mouse.80 healthy female Kunming mice of21 days old were randomly divided into 8 groups:the control group,the estradiol group,the Actinolite group,the Cuscute Senmen group,the Epimedii Foliumetc group,the high,middle and low dose of Huoqi Perfusategroups,10 mice in each group.The first two groups were intragastric administrated with normal saline,the other groups were intragastric administered the extracts of their respective herbs.The drug evolume was 10 ml/kg,per day for 7 consecutive.On the 4th day,0.2 ml of estradiol was injected intraperitoneally once a day for 4 days in the estradiol group,and took sample after the last administration 24 h.The body weight,organ index,uterine horn length,mid uterine diameter,hormone level,receptor gene expression and antioxidant level were studied respectively after treatment withthe Huoqi Perfusate.The results indicated that the middle dose of Huoqi Perfusate,Actinolite,Cuscute Senmen andEpimedii Foliumetc can increase the expression levels of LHR and FSHR,in addition,Actinolitecan increase the levels of LH.The results showed that the content of T-SOD of liverall increased at different degrees in all treatment groups,and the Huoqi perfusion of high dose effect was the best.Additionally,all treatment in our study had the significant effect ofpromote the growth and development of rat uterus,promote the development and maturation of rat follicles,and enhance the antioxidant capacity of rats.2.Effect of the Huoqi Perfusate on Reproductive Function of Female rats with kidney-yang deficiencyThe kidney-yang deficiency rat was established by intramuscular injecting hydrocortisone,Kidney-tonifying pill is used as the control.Before and after treatment with the Huoqi Perfusate and it,s main single herbs body weight,organ index,uterine angle length,diameter of uterus,hormone level and their receptor gene expression,antioxidant levels,blood routine and blood biochemical indicators were detected respectively.The results indicated that the treatment can increase the liver index of rats with kidney-yang deficiency;the high and middle dose of Huoqi Perfusate and Cuscute Senmen,Epimedii Foliumetc can increase the length of uterine horn of rats with kidney-yang deficiency at different degrees.The results showed that the blood level of LH,FSH and E₂ and the expression levels of LHR and FSHR in ovary as well as ER?in uterus all increased at different degrees in all treatment groups.The results showed that Actinolite,Cuscute Senmen and the high and moderate Huoqi Perfusate can increase the content of CAT,in addition,the high dose of Huoqi Perfusate can increase the content of T-SOD of liver.It indicated that each drug delivery group can improve the symptoms of kidney yang deficiency rats.
