Peste des petis ruminants?PPR?is an acute contagious disease of ruminants such as goats,sheep and the like caused by Peste des petis ruminants virus?PPRV?,and its morbidity and mortality Rates are higher.The World Animal Health Organization lists it as a statutory report of the animal epidemic,Our country lists it as class one animal disease rule.On April 3,2014,a suspected epidemic of PPR was reported in Jiajuan Village,YanchangbaoTown,Dingbian County,Y?Lin City.On April 5,the outbreak was confirmed by the National Center for foreign animal disease diagnosis.The relevant departments initiated the contingency plan and disposed of the infected animals with bio-safety.The current PPR epidemic in China emerged:the spread of fast,affecting a wide range,high risks and a grim epidemic situation,which seriously threaten the animal health safety in our country.As a new incoming disease,this disease lack of information on immunization.The actual effect of large-scale vaccineimmunity was concerned by sheep farmers and related fieled persons.For the grim epidemic situation,establishment of RT-PCR detecion methods of PPR,studying of the safety of PPRV caccine,the growth and decline of maternal antibody and the duration of immune antibodies have great significance for the comprehensive prevention and control of PPR.The main contents and results of this study are as follows:1.Through experiment for the establishment of one-step RT-PCR method for detection of the region of Peste des petits ruminants virus N gene.To detect the different material samples from disease foci by using this method.The results show that the method of disease more selective material optional,positive bands detected by sequencing analysis found that strains and strains of Shaanxi genotype was not significantly differen.With high sensitivity and specificity of this method,the concentration of 10-2TCID50/mL could be detected.The detection method for the clinical diagnosis of the disease,improve the accuracy of the fast diagnosis ability and diagnosis of Peste des petits ruminants epidemic in our city.2.2.The experiment with different immunization doses and inoculation methods of PPR showed there was no adverse reactions giving 1 dose and 2 doses by subcutaneous injection or intramuscular injection according to the specification.Test results using SPSS software for chi-square test,P value>0.05,The level of immune antibody was no significant difference,so it is unnecessary to increase the dosage during the non-special circumstances.Antibody was produced after 7days immunization,the quanlification rate of antibody reached more than80%,and it reached the antibody level peaked of 100%qualification rate 14 days later.3.The experiment researched of a number of Chasmere goat of Northern Shaanxi about the safety of PPR vaccine,the growth and decline of maternal antibody,the duration of immune antibodies,and compairing the accuracy of three commonly used serological diagnostic methods.The results showed that the PPAR vaccine was ssfety in clinical application;the average block rate and positive rate of lambs'maternal antibody was reduced after 72-days-old,and it wsa suitable for the first immunization;the mmune duration could reach 67 weeks;the positive rate was significant differnce between colloidal gold diagnostic kit and blocking ELISA.
