The soils were sampled by three-point-sampling method from perennial poplar rhizosphere soil and wood bark in Ning Yang Gao-qiao logging station of Shandong Tai’an.The azotobacter,anti-phosphobacteria,anti-potassium bacteria were separated and screened using cultivated selective medium by the gradient dilution method.Antagonistic bacteria was screened from the soil and wood bark samples,with Botryosphaeria dothidea,Fusarium oxysporum and Fusarium oxysporum as target pathogenic bacteria benzoic acid degradation bacteria were cultivated by mineral medium domesticated uninterruptly.With mensuration biologos of each strains,the strains with higher liveness was selected to study the colonization regularity.The strains which could be colonizated was to be appraisemed and optimized by nutrient medium.Ferment thalli with the best nutrient medium to preparing edaphon repair agent against poplar continuous cropping barrier.And the growth-promoting effect for poplar of the microbial manure was studied.The main results were as follows:1.17 isolates of azotobacter-bacteria,11 isolates of organophosphorus-bacteria,12 isolates of inorganic phosphorus-bacteria,13 isolates of potassium bacteria,54 isolates of antagonism-bacteria,25 isolates of benzoic acid degradation bacteria were isolated from poplar rhizosphere soil;28 isolates of antagonism-bacteria was isolated from wood bark.2.The bioactive measuration of function strains which selected from poplar rhizosphere soil and wood bark showed,the nitrogenase activity of 17 isolates of azotobacter-bacteria were situated in 26.7~229.2 nmol C2H4/hml.And the nitrogenase activity of N-3-6 and N-3-9 were the highest of the total.The muddy circle diameter in egg plates of 11 piece of inorganic phosphorus-bacteria were 2.24~2.57 cm after 24h,3.01 cm and 4.25 cm after 72h.And the solutions organophosphorus activity of PY-2-6 and PY-3-7 were the highest of the total.The transparent circle diameter in phosphorite plates of 12 piece of anti-inorganic phosphorus bacteria were 1.17~1.41 cm after 24h,1.24~1.65 cm after 72h.And the solutions inorganic phosphorus activity of PW-2-7 and PW-3-11 were the highest of the total.The K+content in supernate of 13 piece of benzoic acid degradation bacteria was increased 10.10%~65.77%compared with comparison which contcted blancing bacteria.The result of K-2-2、K-2-12 and K-3-3 were best of the total.The bacteriostasis width of rhizosphere antisepsis T1-24,T5-1,T6-1 and T6-6 was larger than 5.00 mm.The of benzoic acid degradation rate of 25 piece of benzoic acid degradation bacteria were situated in 6.96%~80.23%.And the antagonism effect of bark endophyte N3-23 and N6-34 were the best of the total.The bacteriostasis width was larger than 5.00 mm.3.Rhizosphere bacteria and endogenous bacteria use resistance tag to study the colonization ability.The results show that,K-1-8、K-2-2、PW-3-11、PY-2-6、PY-3-7、N-3-6、N-3-9、BJS-1-3、N3-23 and N6-34 were colonization stably.4.The results mutual antagonism showed,K-2-2、N-3-9 antagonistic activity against N3-23,N-3-9、PY-3-7 antagonistic activity against N6-34.5.The results of thalli shape and bacterial colony,physiology and biochemistry indicator analysis,16S rRNA analytic result show that,N-3-6 belonged to Stenotrophomonas sp,PY-2-6 belonged to Chryseobacterium sp.,PW-3-11、K-1-8、BJS-1-3、N3-23 and N6-34 belonged to bacillus sp..6.The orthogonal design results of optimization medium shows that Medium of all strains were different.Formula as follows:The medium of PW-3-11:glucose 20g,indian meal 20g,soybean powde 40g,(NH4)2SO41.0g,KH2PO40.5g,CaCO3 1.5g,water 1000mL;The medium of K-1-8:glucose lO.Og,indian meal 20.0g,soybean powde 20.0g,(NH4)2SO4 15g,KH2PO4 1.5g,CaCO3 0.5g,water 1000mL;The medium of N-3-6:glucose 5.0g,indian meal 40.0g,soybean powde20.0g,(NH4)2SO4 1.0g,KH2PO40.5g,CaCO3 0.5g,water 1000mL;The medium of BJS-1-3:glucose 10.0g,indian meal 40.0g,soybean powde 10.0g,(NH4)2SO4 1.0g,KH2PO41.5g,CaCO3 1.0g,water 1000mL;The medium of N3-23:glucose 20.0g,indian meal 20.0g,soybean powde 40.0g,(NH4)2SO4 1.0g,KH2PO40.5g,CaCO31.5g,water 1000mL;The medium of N6-34:glucose 20.0g,indian meal 10.0g,soybean powde 20.0g,(NH4)2SO40.5g,KH2PO41.5g,CaCO31.0g,water 1000mL;7.The induced systemic resistance results of endophyte show that,the N3-23 and N6-34 could increase the enzyme activity of POD and SOD,observably.8.0.5 kg piece tub,microbial manure were added to the flower with 1 year-old poplar young plants using the method of hole application.And the biomass of poplar young plants were mensured.The result showed,young sapling stem diameter and shoot length increased significantly compared with control group.
