Research On The Composition And Distribution Of Endophytic Bacteria And Fungi In Acanthopanax Senticosus Of Northeast China

Abstract/Summary:
Eleutherococcus senticosus is one of the most important Chinese herbal medicines in Northeast China,and its root,stem and leaf can be used as medicine.The present study of Acanthopanax endophytic bacteria are mainly two aspects of bacteria identification and biological activity of endophytic bacteria,studies show that isolated from Acanthopanax senticosus endophyte has various biological functions,such as broad-spectrum antibacterial activity,can significantly improve the biological activity of Acanthopanax senticosus metabolites and so on.However,little is known OTU the analysis of microbial community in Acanthopanax senticosus.This study used high-throughput sequencing of Acanthopanax endophytic bacterial community analysis,experiment of Acanthopanax senticosus collected from Heilongjiang province in Maoershan,Heilongjiang Province,Xingan small Ling Jilin province Changbai Mountain three geographical regions,respectively in three regions of acanthopanax root samples,after removal of surface microbial DNA extraction,were detected in the bacteria and then in the classification of different fungal sequences,three unit area of acanthopanax root community diversity.In this paper,the endophytic bacteria communities in the root,stem and fruit of the same Acanthopanax senticosus in the Xingan area were analyzed.The study found that in Changbai Mountain,Maoershan,Xingan mountains in three regions of Acanthopanax endophytic fungi in the roots of Sordariomycetes,Tremellomycetes,Dothideomycetes is the main phylum of fungi including Fibulorhizoctonia,Phialocephala,and Russula is a genus of fungi in Xiao Xing’an Ling region of acanthopanax root unique endophytic fungi,indicating that acanthopanax root thorn in different regions endophytic bacteria had significant difference.The same strain of Acanthopanax senticosus in Xiao Xing’an Ling stem Sordariomycetes phylum of fungi accounted for 79.80% of the proportion,in the fruit samples found in the Tremellomycetes phylum of fungi occupy the entire root fungi 51.16%,due to the distribution of relatively stable environment factors of endophytic fungi in uniform.It was found that although the number of fungi in the same area was similar to that of fungi,the proportion of fungi in different parts was different,because the microenvironment of each part was different.In the three regions in the Erwinia study found that acanthopanax root thorn endophytic bacterial communities as a plant pathogen in Maoershan and small Xingan mountain area of acanthopanax root is widely distributed,and the content is stable,but the Changbai Mountain area of acanthopanax root Erwinia rarely,this the Changbai Mountain area of Acanthopanax plants on Erwinia infection with immune ability,which has important guiding significance for the artificial cultivation of Acanthopanax senticosus.On the same line in different parts of Acanthopanax senticosus bacterial communities found in the genus Enterobacteriaceae are abundant in fruits,roots were 29.7% in the stem was only 6.8%,indicating that Enterobacteriaceae is possible in the process of reproduction of Acanthopanax senticosus with genetic to the next generation.The correlation analysis of the climatic characteristics of the three regions and the distribution of endophytic bacteria in relation to carry on the analysis,found that the influence of three main factors of area thorn bacteria distribution difference in altitude is five students;the small Xingan mountain area Acanthopanax bacteria analysis found that the students do not have parts,small Xingan mountain area the same strain thorn main factors different parts of Acanthopanax endophyte is light intensity difference.This shows that there is correlation between the distribution and location of Acanthopanax thorn endophytes in climate,the study found that thorns have bacterial distribution difference and acanthopanax place of the climate in the distribution and high correlation of Acanthopanax,endophytic fungi under various conditions,so the acanthopanax distribution of endophytic bacteria can reflect the acanthopanax local climate environment.This paper presents a new method of isolation,samples were sequenced to get basic information about the corresponding microbial community of bacteria,it can go to the goal of screening of endophytic bacteria,so this study were separated by sequence comparison showed that Pantoea cypripedii Pseudomonas marginalis,Pseudomonas sp.,three strains of bacteria in sequencing analysis of Acanthopanax senticosus in endophyte,acanthopanax isolated Phialocephala fortinii,Gibberellaavenacea,Hypocrea.virens three fungi are appeared in the analysis of 454 highthroughput sequencing in fungi.It shows that the idea is feasible.
