At present,the role of MHC molecules has been widely concerned in human immunoregulation,organ transplantation and autoimmune diseases.In this study,blood samples and liver tissues were collected from 15 Landrace pigs in a pig farm of Changchun and the blood genome and tissue RNA were extracted respectively.After successfully synthesizing the primers,the second exon of SLAII-DRB1 was amplified by PCR obtaining the target genes which were allelic discriminated by sequence-based typing and the nucleotide polymorphism was analyzed.118 valid sequences whose length was 245bp(one of which was 246bp)were obtained by editing and comparing the acquired 225 nucleotide sequences using the relevant biological software.After the sequences analysis,43 allelotypes were obtained.The allele with the highest distribution frequency was found in all individuals,and many alleles were distributed in only one individual.In the analysis of the second exon genetic polymorphism,the SLAII-DRB1 gene was highly polymorphic.The swine serum antibody was also detected by using Streptococcus suis type 2 Antibody ELISA Detection Kit.And the positive rate of antibody was 73.3%.High-throughput sequencing technology is also known as the next generation sequencing,with high throughput,micro,low cost and so on.In this study,the Illumina Hiseq2500 sequencing system was used to detect the pathogenic microbial diversity in pig blood.The 16 SrDNA gene library from 12 pig blood samples was constructed by using primers of 16 SrDNA V3 + V4 region,and the products were sequenced by Hiseq2500 after PCR amplification and purification.PE reads of all samples were filtered to Clean Tags which were used to analyze OTU and calculate alpha diversity index and draw rarefaction curve.The results showed that the quantity of samples and data analysis had reached enough depth and the microbial diversity in the blood samples was generally high.The microorganism in 12 samples was classified according to phylum and genus profiling.And the distribution of Streptococcus in Sus3,Sus6 and Sus13 individuals was found to be significantly higher than other samples.According to above results,the special alleles in Sus3,Sus6 and Sus13 individuals could be the SLA resistance alleles associated with Streptococcus suis 2 infection according to above results. |
