Abstract/Summary:
Streptococcus suis is a general term for disease of pigs caused bystreptococcal bacteria. A very wide distribution range of the disease has occurredaround the world. The Netherlands, United Kingdom, the United States and othercountries around the world have reported the Streptococcus Suis since confirmed inearly1950s.The disease seriously affected the development of the every national pigindustry, especially for high-density pig farms, because Streptococcus Suis havemany serotypes and complexity of the antigenic structure. The disease has causedtremendous economic losses to the swine industry and received more and moreattention. In recent years, China’s Streptococcus Suis becomes more and moreserious in addition to Tibet and other regions where have not yet found the disease.The disease have different levels of occurrence and epidemics in most of theprovinces, municipalities, autonomous regions, and cause employees to infectionand death, which serious harm human public health security. So build a new, fastand accurate Streptococcus Suis detection method is particularly important.Gene chip or microarray, the combination of biology and microelectronicstechnology, is a kind of new micro-device. Through the match of mutation probesand sequences, we can detect the mutation quickly, lightly and accurately. Thedetection can effectively instruct animal diseases diagnosis. It is composed of DNAarrays immobilized on solid supports. Because of its many virtues, such as rapid,sensitive and accurate, gene chip plays an important role in the study of automateddisease detection tools.The study utilized microbial science, Serological, PCR identification and genesequencing were performed to genotyping identification the molecular epidemiology of the Streptococcus Suis, tried to acquire new efficient methods forpreventing and treating Streptococcus Suis better and better.In this study, the type2standard strains of specific probes produced genechips, the captor-DNA fragments, which were reamplified from Streptococcus Suisserotype2, were spotted on a glass-bound microarray. Cy3-labeled fluorescent PCRproduction was hybridized to the captor-DNA on microarray. At the same time, westudied the effects of the different concentration of the captor-DNA andhybridization time on the detecting result, confirmed the concentration ofcaptor-DNA is5μm/L,42℃water bath temperature bath30min, to get a betterhybridization signal.The results show that microarray diagnosis method ofStreptococcus suis serotype2established in this research can successfully verify thespecific sequence of the Streptococcus suis serotype2, the signal of the fluorescenceis clear, the specialty is high. In fact it is an efficient diagnosis method. Not only fora variety of infectious diseases in the same piece of pig lay the foundation fortesting the chip, but also for other animal diseases were detected by gene chipmethod provides a theoretical basis.To sum up, this study successfully established a diagnosis technique platformof microarray and solved the difficult problem for Streptococcus Suis serotype2,with afford a theoretical base for prevention and diagnosis of animal’s epidemicdiseases, control and annihilate epidemic.
