Metronidazole,a kind of antibiotics,is the first generation of the 5'-nitroimidazole,antibacterial mechanism of anaerobic bacteria by acting on DNA transcription and translation process,inhibiting the synthesis of bacterial DNA,thereby affecting bacterial replication,growth,to achieve antibacterial effect.It widely used in clinical diseases such as subcutis,gastrointestinal tract and soft tissue infections caused by anaerobic bacteria.The experiment used the formulation of metronidazole injection through intravenous with a rapid rate and completely absorbed,then widely distributed in various tissues and body fluids.And it can through the blood-brain barrier.The effective concentration of the drug can be present in saliva,bile,milk,urine,pus,and cerebrospinal fluid.Its concentration is high within tissues and it has apparently post-antibacterial effect.This article from the following aspects study MTZ Injection dogs clinical safety and pharmacodynamics,and thus provide a reference for clinical medication pet.Experiment 1 The study of metronidazole injection on clinical safety in dogsAccording to the Ministry of Agriculture bulletin(No.1425)"Safety testing guidelines of pet's drugs on the target animal" to valuates the clinical safety of metronidazole injection in dogs.The test selected 24 healthy adult dogs were randomly divided into four groups,six dogs in each group,half male and half female.According to the clinical recommended dose of metronidazole(15mg·kg-1)as the basis,respectively set the 5 times clinical recommended therapeutic dose group(75mg·kg-1),3 times clinical recommended therapeutic dose group(45mg·kg-1),the clinical recommended therapeutic dose group(15mg·kg-1)and the control group(0 mg·kg-1).Vein injection for 14 consecutive days,at the trial periods,by comparing the changes of clinical symptoms of dogs,when they were at the 1st day before the test,the 4th day after administration,the 7th day after administration,the 14th day after administration and the 7st day after ending the test,such as blood CBC indicators,blood biochemical indicators,urine test indicators and histopathology to evaluate the safety of metronidazole injection.The results showed that:compared with the control group,there is no significant difference(P>0.05)between the clinical recommended therapeutic dose group,3 times the clinical recommended therapeutic dose group and 5 times the clinical recommended therapeutic dose group after 14th day administration.Experiment 2 Establish about modeling subcutaneous soft tissue anaerobic infections on dogsFor studying metronidazole injection curative effect about subcutaneous soft tissues of dogs which were affected by anaerobic bacterium,we must firstly establish a model of subcutaneous soft tissue anaerobic infections on dogs.Methods:Check the trial dogs,body condition to determine all trial dogs are healthy.Then anaerobic bacterium were injected into the subcutaneous.Next,we observed the clinical symptoms,tracking by ultrasound probe,get the local aspiration fluid by fine needles to make a smear staining,bacterial isolation and biochemical identification.Results revealed:Local abscess,swelling and pain.Ultrasound examination shown that diffuse hypoechoic or irregular moderate to hyperechoic as well as irregular discrete anechoic,while the margin of the lesions is not clear and aggressive.Track and detect the lesions we found them and normal subcutaneous tissue gradually infiltration into each other.Smear staining shown a kind of bacillus which were stained dark in large blunt ends with non-staining in the middle;Biochemical identified them as the model bacterium.Experiment 3 The efficacy of metronidazole injection for subcutaneous soft tissue anaerobic injections in dogsSelect successful modeling 18 dogs were randomly divided into three groups of six,model control group,metronidazole injection group and Ceftiofur sodium group,were Vein injection treated by saline,metronidazole with clinical recommended dose(15mg·kg-1)and Ceftiofur sodium with clinical recommended dose(5mg·kg-1)for 7 d,respectively,bid.The results showed that both metronidazole and Ceftiofur sodium were sensitive against anaerobes.Compare to Ceftiofur sodium,metronidazole showed stronger inhibitory effect.After administration of metronidazole,abscess increasingly affected area back to normal,continuous medication 7 d can be basically recovered,and the recovery rate and the overall situation is slightly better than the Ceftiofur sodium group.Metronidazole group and model group detection indicators presented in discontinued after 7 d with significant difference(P<0.05),showed metronidazole injection is effective in treating anaerobes infections in dogs,rational its recommended dosage.
